In recent years the research-development and application of probiotics have been vigorously promoting. They can replace antibiotics and effectively reduce the serious threat of antibiotics abusing to our livestock industry. Because of the diversity and changes of intestinal flora, mammalian body development can be reacted in health and disease. This study was designed isolate a butyric acid-producing bacteria, which is belong to Clostridium butyricum, acid and bile tolerant, adapt gastrointestinal environment well, from fresh pig manure. In order to provide a theoretical basis for the study of probiotic and Clostridium butyricum complex preparations, we discussed and researched the effects of intragastric administration with this probiotic strain and Lactobacillus salivarius in mice.In this study, the separation of bacteria, culture, purification, staining, microscopy, etc. to give the suspected strain. And combined with 16S rDNA sequence comparison and identification of bacteria Bergey’s Manual of their identification. We also did some research on this strain for its growth characteristics. Some researches have been done to observe the effect of growth performance, diarrhea, mortality, blood indicators, serum biochemical indexes and intestinal sections in mice by this strain and Lactobacillus salivarius gavage and studied the changes of intestinal flora by DGGE method.The results showed that a butyric acid producing bacteria, which grow well in mammalian digestive tract, was isolated from fresh pig manure. There was no effect on growth performance, diarrhea, mortality, blood indicators in a short period of time by Clostridium butyricum and Lactobacillus salivarius gavage in mice(P>0.05). Clostridium butyric acid have a significant effect on serum biochemical indexes and enhances antioxidant(P<0.05).Lactobacillus salivarius showed no effect(P>0.05), and the mixed group decreased serum triglyceride. Both Clostridium butyric acid and Lactobacillus salivarius have significant effect on increasing the villi, reducing crypt depth, expanding the contact area of the intestine and thickening colonic mucosa thickness(P<0.05). But no significant effect on cecal(P>0.05).This study showed the dominant intestinal flora are Bacteroides and Firmicutes of mice by DGGE and stripe recovered cloned and sequenced. Clostridium butyricum and Lactobacillus salivarius can Promote Bacteroides and Firmicutes in intestinal colonization. But the diversity index and the equilibrium degree of intestinal flora did not change significantly after Clostridium butyricum and Lactobacillus salivarius gavage. And the abundance decrease.
