This paper mainly studied the methods of preparation for immobilized cellulase, investigated the conditions of the experimental terms and immobilized enzyme column preparation for making corn germ oil and observed related components microscopic structure changes in this processing.The experimental parameters were chose for experimental immobilized enzyme column preparation for making corn germ oil.Further more, antioxidant activity of protein hydrolysate was detected, the main results were as follows:1.In the processing of aqueous enzymatic method extraction of corn germ oil, the effect of cellulase enzymolysis was good, the best technological parameters were: the enzyme adding amount was 997.6 U/g, enzyme pH was 9, material liquid ratio was 1:6, extraction temperature was 46 ℃, the extraction time was 4.3 h.Oil extraction rate could reach 61.26%.Total fatty acids in corn germ oil content was above 85%, the total relative content of oleic acid and linoleic acid were as much as 83%.2.Based on the response surface and single factor experiments, the best technology of sodium alginate immobilized cellulase was: free enzyme amount was 10000 U/g, the concentration of sodium alginate was 2.8%, CaCl2 concentration was 4.2%, the immobilization time was 4.2 h, the final enzyme activity recovery rate was 72.85%.3.Based on single factor and response surface experiments, the best technology of chitosan immobilized cellulase was: the concentration of chitosan was 2.05%, glutaraldehyde concentration was 0.51%, the immobilization time was 14.20 h, crosslinking time was 3.10 h,the final enzyme activity recovery rate was 70.67%.4.Under the protection of the cellulose enzyme immobilization carrier, it showed a greater heat resistance and thermal stability.The highest condition temperature could reach 60 ℃,the enzyme still got the active, the recycle time of the immobilized enzyme could reach 5 times, and the initial relative enzyme activity maintain 60% in 20 days.5.The best conditions for extraction of corn germ oil by immobilized cellulase column reactor optimum technological were: enzyme solution temperature was 50 ℃, pH value was 9, the flow rate was 5 mL/min, and solid-liquid ratio was 1:8, the processing time was 8 h, oil extraction rate could reach 50%.6.Corn germ of the corn germ protein was hydrolyzed protein composition and amino acid composition similar to that of corn germ protein hydrolysate amino acid content of 32.2% was 2.6 times that of corn germ protein isolates of 12.5%. When corn germ protein hydrolysate concentration was 15 mg/mL, the reducing power can reach 1.542, the clearance rate of hydroxyl free radicals and DPPH were 59.31%, 53.11% respectively.Corn germ protein hydrolysate showed good antioxidant activity, and functional properties of corn germ protein hydrolysate was better than that of protein isolated.
