Pyrazosulfuron-ethyl belongs to the sulfonylurea herbicide, and it is mainly used in rice field. With the large-scale application of the pyrazosulfuron-ethyl, it becomes an increasing-concern problem beause of the bad effect on the agricultural production and the environment. So it is necessary to study on the mechanisms of pyrazosulfuron-ethyl. In this study, proteomics was used to gain further insights into the adaption mechanisms of Rhodopseudomonas palustris PSB-S, and the main results were as follows:The Rhodopseudomonas palustris PSB-S was selected by relative resistance measuring and used as material in following experiment.Comparative proteomics was used to gain further insights into the adaption mechanisms of Rhodopseudomonas palustris PSB-S. In order to identify differentially expressed proteins,2-D electrophoresis and MALDI-TOF/TOF-MS was carried out, and 56 differently expressed proteins were successfully identified. Then the enriched functional categories and the biological metabolic pathways of the differentially expressed proteins were analyzed. The results showed major variations in the composition of outer membrane proteins and receptor or transport proteins.In addition, several proteins involved in protein synthesis, folding and stabilization were up-regulated, some proteins related to energy production were down-regulated.The respective gene expression patterns of several identified proteins were evaluated by quantitative real-time PCR and shown to consistently correlate with those deduced from the proteomic study.The effect of pyrazosulfuron-ethyl on Rhodopseudomonas palustris PSB-S morphology was evaluated by scanning electron microscope and transmission electron microscope, and shown to consistently correlate with those deduced from the proteomic study too.In summary, it is the first time to reveal the adaption mechanisms of Rhodopseudomonas palustris PSB-S by the proteomics.And it provides new genetic resources to creat pyrazosulfuron-ethyl resistant crops and provides new resources for bioremediation of pyrazosulfuron-ethyl pollution.
