Persistent organic pollutants which are harmful for the ecological environment and human health bring a range of environmental problems and seriously threaten the sustainable development of human society. Such substances are persistent, bioaccumulative, relatively stable, and easily adsorbed on the soil and particles. In May 2009, the United Nations environment Programme formally list four polybrominated diphenyl ethers, five polybrominated diphenyl ethers, six polybrominated diphenyl ether and seven polybrominated diphenyl ethers in the Stockholm Convention because of its potential great harm. Reports on PBDEs of toxicology, risk assessment, elimination is relatively small, and detection is also largely dependent on the traditional instrument analysis, so a more rapid and sensitive detection of PBDEs will contribute significantly to these research. Compared to instrumental analysis, immunoassay based on antibody and antigen-specific immune response, is rapid, sensitive, and low cost can do large quantities of goods field detection, so immunoassay development of PBDEs detection is of great significance.The high specific and affinitive antibodies are the core of the immunoassay rea gent, but small molecules such as PBDEs have no immunogenicity. To crack the situation, researchers couple the PBDEs to a carrier protein to induce animal to produce antibodies. Unfortunately PBDEs does not contain the reactive groups for coupling, therefore it is especially critical to design and synthesize the haptens that retain the parent structure characteristics and contain a reactive group. Based on these, the papers do the following work:Firstly, the synthesis method, raw materials and connection arm access point are determined by analysis of PBDEs and its derivatives. We also explore the experimental conditions to optimize the synthesis method.Secondly, in order to increase the possibility of getting high specific and affinitive antibodies, we design and synthesize tetrabromodiphenyl, penta-and six-brominated diphenyl ether carboxylic acids immune hapten according to basic design requirementsThirdly, in order to improve the detection sensitivity by using a heterologous detection system, two kind of PBDEs carboxylic acids with different length connecting arm are designed and synthesized.
