| Cinnarizine is a Ca2+ channel blocker that plays an expansion role on vascular smooth muscle and make obvious improvement on cerebral circulation and peripheral circulation. The clinical performances of Cinnarizine are mainly used on the treatment of cerebral thrombosis, cerebral arteriosclerosis and other diseases. The traditional preparation is large and frequent dose and easy to cause the body side reaction. In terms of the disadvantages of traditional preparation, the research adopts emulsion solvent evaporation method to make uniform film and hybrid film Cinnarizine microcapsules preparation. This research analyses effect factors including surface morphology, particle size, yield, encapsulation efficiency, and drug release rate in vitro by orthogonal test in order to improve the bioavailability of Cinnarizine, reduce drug dosage and reaction and provide certain theoretical basis for practical application of drug microcapsule.The results of research show that the yield of EC drug microcapsule is best when the concentration of EC, SDS, PVA are 4.0%, 0.05% and 1.0% g? m L-1 respectively,oil-water ratio with 1:8 and CIN-PS ratio with 1:12. When the concentration of PS, SDS,PVA are 1.0%, 0.15% and 1.0% g?mL-1 respectively, oil-water ratio with 1:10, CIN-PS ratio with 1:10, the yield of PS drug microcapsule is best. When the concentration of EC,SDS, PVA are 4.0%, 0.10% and 2.0% g?mL-1 respectively, the ratio of CIN to EC with 1:14 and oil-water ratio with 1:8, the encapsulation efficiency of EC drug microcapsules is best. When the concentration of PS, SDS, PVA are 1.5%, 0.15% and2.0% g?mL-1 respectively, the ratio of CIN to PS with 1:16 and oil-water ratio with 1:6,the encapsulation efficiency of PS drug microcapsules is best. According to research,for different proportion of EC and PS mixed drug microcapsule membrane, the yield of drug microcapsule increases with the increase of EC content in the membrane material.The result of cumulative release shows that drug releases at a constant speed and keepsmore than 8 hours. |
