Infant formula is a kind of powered food which designed for the nutritional needs of infants and young children at different growth stages. With the advent of free trade era, the infant formula market is highly competitive, and there have been some pseudo products in the market, causing widespread concern in society. Therefore, a rapid and simple identification of infant milk powder quality is very important. In this paper, optimizing the chromatographic conditions by means of RP-HPLC, establishing a brand of infant formula fingerprints and comparing different brands of infant formula, which provide an effective testing methods for the infant milk powder identification and protection of brand. Specific results are as follows:(1)Determining chromatographic detection conditions based on the separation characteristics of infant formula protein chromatography. RP-HCLP analysis was performed on a Shimadzu C8 column(250mm × 4.6mm, 5 μm), mobile phase A was 0.1% aqueous trifluoroacetic acid in water, mobile phase B was 0.1% trifluoroacetic acid in acetonitrile,using gradient elution, each sample analysis time is 45 min. Selecting 214 nm for detecting wavelength, the flow rate was 0.5m L/min, the injection volume was 20μL.(2)Application of RP-HPLC method analysis of two series of infant formula from YinQiao. Common peaks were confirmed, there were five main peaks. Good reproducibility,stability and precision. The results showed that stage 1, 2 and 3 milk powder maps from A series and B series have respectively overall profile similarity, in line with common fingerprinting.(3)This test conducted by HPLC method, being applied for different brands of infant formula protein, including brand Erie、Firmus、Beingmate, contrasting with the infant formula from B series of YinQiao. Comparing chromatograms from different species of infant formula through the retention time, height, area, to differentiate the brand Erie、Firmus、Beingmate infant milk powder.(4)Four types of infant formula and two series of YinQiao infant formula were differentiated by RP-HPLC fingerprints combined with PCA. The three brands, Firmus, Erie,Beingmate, do not correspond with the letters C, D, E represents. The cumulative variance contribution rate of two main components eigenvectors was over 90%. The PC1 can distinguish between brand YinQiao and D、E、C infant formula, the brand D, E samples are separated from brand C on PC2. Infant formula milk powder from A, B series of YinQiao canbe distinguished on the whole.
