In this article, with casein as raw materials, casein enzymatic hydrolysate was prepared by Alkaline protease hydrolysis.Casein phosphopeptides was extracted from casein enzymatic hydrolysate.And then casein phosphopeptides was chelated with zinc. We made optimum chelating parameters clear by response surface methodology which based on the single-factor experiments.Casein phosphopeptides chelating zinc as a new zinc nutritional supplements, it is compared with zinc sulfate and zinc glycinate. The effect of zinc sulfate, zinc glycinate and casein phosphopeptides chelating zinc on the early growth of mice and the effect of zinc supplementation were analyzed.Lay a foundation for the next test. 1 Response surface method optimization casein phosphopeptides chelating zincEffects of zinc source type, concentration of casein enzymatic hydrolysate,. mass ratio of casein phosphopeptides to zinc, reaction pH value, temperature and time on chelation of casein phosphopeptides with zinc were analyzed.Results showed that zinc in form of ZnSO4·7H2O presented a higher chelating capacity with casein phosphopeptides than that in forms of Zn(Ac)2 and ZnCl2.Better chelation effect was obtained under the conditions of mass ratio of casein phosphopeptides to Zn2+ 4:1, pH 7.0, reaction temperature 60℃, and reaction time 80 min. In this condition, the chelation rate was 92.95%. 2 Effects of different zinc source on the early growth of mice and the effect of zinc supplementation in miceForty male KM mice were randomly divided into four groups. The control was fed on basal diet, zinc sulfate group, zinc glycinate group and casein phosphopeptides chelating zinc group were fed on the diets supplemented with zinc sulfate, zinc glycinate, casein phosphopeptides chelating zinc at 40 mg/kg. Determination of mice body weight, serum zinc content, serum AKP activity. The results showed that adding different zinc source can effectively improve the mice body weight, serum zinc content,serum AKP activity. The mice body weight, serum zinc content, serum AKP activity in the treatment of casein phosphopeptides chelating zinc significantly higher than that of zinc sulfate group, and slightly higher than the zinc glycinate group. The effect of zinc supplementation of casein phosphopeptides chelating zinc is superior to zinc sulfate and slightly better than zinc glycine.
