In this paper,using Inner mengolia Xilinguole Sunit sheep cartilage as raw matierial prepared the collegan by pepsin and obtained collegan peptides after futher hydrolysis.And physicochemical properties, antioxidant activity of collegan peptide in vitro and vivo was investigated,respectively.1.Comparative study of physicochemical characteristics on collegan and collegan peptide of Sheep Cartilage was annalyzed.The results showed that the solubility, water absorption, oil absorption, emulsifying and foaming properties of collegan peptides are better than collagen,solubility and foaming properties significantly higher than collegan.2.Using centrifugal ultrafiltration tube separated collagen peptide throuh membrane and obtained 3 kinds of sheep cartilage collagen peptide fractions,which molecular weigh 5kDa-10kDa,3kDa-5kDa and less than 3kDa,respectively.different fractions’ antioxidant activity in vitro was studied. The results show that antioxidant activity of collagen peptide with different molecular weight ranges was different. With the increase of the concentration of antioxidant activity increased, hydroxyl radical(OH)scavenging acvtivity of hydrolysate reached 96.71%,was significantly higher than Vc(P<0.05). superoxide anion free radical (O2-·)Scavenging activity and Reducing abilityof collegan peptide which molecular weight was 3kDa-5kDa was strongest,but was not better than that of Vc and the difference was significant(P<0.05).3.The different dose of bone collagen peptide mixed with soybean oil and lard oil system to study its oxidation inhibition ability of different oil system.The results showed that sheep cartilage collagen peptide can extend the lipid oxidation induction period,and the higher the additive amount,the lower the peroxide value,the stronger the antioxidant ability.4. The mice were divided into normal group, D-galactose model group, and the high and low dose group studied the sheep cartilage collagen peptide antioxidant activity in vivo. The results showed that,compared with normal group, D-galactose model group’s CAT, GSH-Px and SOD activity in the mice liver tissue significantly decreased (P< 0.01), MDA content was significantly increased (P< 0.01),and the content of hydroxyproline in the skin significantly reduced (P< 0.01).High dose collegan peptides can significantly enhanced the CAT, GSH-Px and SOD activity of mice liver tissue which induced by D-galactose (P< 0.01).And can decreased the content of MDA (P< 0.01), increased the content of hydroxyproline in the skin of mice,significantly (P< 0.01).
