Quantum dots, zero-dimension nano material, possesses the basic effect of nano materials, and has been widely used in many areas, such as biology, chemistry, physics, medicine and materials science. Quantum dots synthesized in aqueous phase has biocompatibility, and can be directly applied to biological systems. Quantum dots with excellent properties are synthesized with thioglycollic acid as stabilizer, and application in biological systems. The research contents of paper are as follows:1. Interaction of CdSe quantum dots and ovalbumin (OVA). We used thioglycollic acid as stabilizer to synthesize CdSe quantum dots in aqueous solution directly, and CdSe quantum dots was characterized by ultraviolet spectrum and fluorescence spectrum. Ultraviolet spectrum scans findings: CdSe quantum dots had characteristic absorption peak at 460 nm; fluorescence spectrum showed that the CdSe quantum dots had a single emission peak at 650 nm. We studied interactions of CdSe quantum dots and ovalbumin (OVA) via ultraviolet-visible absorption spectroscopy and fluorescence spectroscopy. The results showed that CdSe quantum dots can quench fluorescence from OVA, in order to determine the type of quenching mechanism, fluorescence spectra was determinated at 25℃ and 30℃ and 37℃, the results showed that the type of quenching belongs to static quenching mechanism. Synchronous fluorescence results showed that the spike of characteristic peak of tyro sine and tryptophan did not change, and explain that the existence of CdSe quantum dots have no effect on the microenvironment of OVA molecule.2. Research of effect of CdSe quantum dots for cell. We studied the influence of CdSe quantum dots on PC-12 cell survival rate and the impact on human peripheral blood lymphocyte micronucleus rate in vitro. The results showed that the low concentration of CdSe less impact on the cell survival rate, and high concentration of CdSe make the cell survival rate decline obviously. The micronucleus test showed that the existence of CdSe quantum dots under trial conditions had no significant effect on the growth of lymphocytes, but cell micronucleus rate increased, found no chromosome aberration, explained that CdSe quantum dots on the lymphatic less affected in the process of cell division.3. Biological effects study of CdSe quantum dots in vivo. We researched the influence on the growth of mice after injecting CdSe quantum dots, the results showed that the liver, spleen and kidney of CdSe treatment group was obviously enlarged, fresh weight increased; we thought that the CdSe quantum dots is harmful to liver, spleen, kidney and other organs in mice and it is toxicological effect on mice. Besides, we also explore the immunological effect of CdSe quantum dots by ELISA experiments. Besides, we measured serum concentration of Cd2+ and Se2- of CdSe treatment group, the results showed that the serum concentrations of Cd2+ and Se2- were higher than control.
