Thuja, has antibacterial, anti-oxidation, anti-diarrhea, anti-tumor and another a s eries of biological activity. In this paper, the natural essential oil abstracted from th uja was the main experimental materials and the object of study.Firstly, the antibact erial activity of thuja essential oil, on Escherichia coil, Staphylococcus aureus, Aspe rgillus niger, Rhizopus, Cladosporium spp and Aspergillus oryzae, which was tested by filtering paper method, was researched as well as the Minimum inhibitory conce ntration (MIC) on these microorganisms were measured by tube dilution method. Th en, the stability of antibacterial activity in different temperature and ultraviolet irradi ation was measured. After that, the antibacterial mechanism was discussed based on the ingredients of essential oil. Secondly, to investigate the properties of thuja oil deep, and to provide direction for the latter product development, the thuja essential oil was mixed with other natural essential oil, the antibacterial activity of the diffe rent formulas were tested by tube dilution method. Finally, with the gelatin and chi tosan as wall material, the thuja essential oil was made into microcapsules by com plex-coacervation. The optimum parameters were researched by orthogonal test and single factor experiment, and the main properties of the product were measured and evaluated at last.The results showed that:1. The antibacterial effect of thuja essential oil on Cladosporium spp, Rhizopu s, Aspergillus niger, Aspergillus oryzae, Escherichia coli, Staphylococcus aureus wer e effective. The MIC were 4%,2%,3%,3%,3%,2% respectively.2. In the condition of low temperature, high temperature treatment and ultravio let irradiation, the thuja essential oil had great stability.3. The ingredients of thuja essential oil mainly were terpenoids, including Japa nese thuja, a-cedar alcohol, Flower arborvitae; etc.The main mechanism of its antib acterial is that terpenes affect respiration of the microorganism.4. In 8 kinds of common plant essential oil (peppermint oil, tea tree oil, rose mary essential oil, eucalyptus oil, geranium essential oil, lemon essential oil, clove essential oil, rose essential oil), clove and eucalyptus essential oil to the selected b acteria inhibition effect is significantly effective than other essential oil. The clove essential oil to the tested microorganisms, the MIC ranged between 0.25% to 1%, a nd the eucalyptus essential oil ranged between 4% to 6%.5. The antibacterial effect of the compound essential oil (clove and thuja esse ntial oil) on Cladosporium spp, Rhizopus, Aspergillus niger, Aspergillus oryzae, Esch erichia coli, Staphylococcus aureus were effective, and the MIC respectively were 0. 4%,0.2%,0.4%,0.8%,1.2%,1.2%. The Fractional inhibitory concentration (FIC) i ndices were respectively synergistic effect, synergistic effect, additive effect, additive effect, additive effect and additive effect. The antibacterial effect of the compound essential oil (eucalyptus and thuja essential oil) on Cladosporium spp, Rhizopus, A spergillus niger, Aspergillus oryzae, Escherichia coli, Staphylococcus aureus were eff ective, and the MIC respectively were 4%,2%,4%,4%,4%,2%. The Fractional i nhibitory concentration (FIC) indices were respectively additive effect, additive effec t, independent effect, independent effect additive effect, and additive effect. The anti bacterial effect of the compoundessential oil (clove, eucalyptus and thuja essential o il) on Cladosporium spp, Rhizopus, Aspergillus niger, Aspergillus oryzae, Escherichi a coli, Staphylococcus aureus were effective, and the MIC respectively were 2.4%, 0.6%,1.2%,1.2%,2.4%,1.2%. The Fractional inhibitory concentration (FIC) indice s were respectively additive effect, additive effect, independent effect, additive effect, independent effect, and additive effect.6. Using the complex-coacervation to produce thuja essential oil microcapsule. The optimum parameters which were researched by orthogonal test and single factor experiment were that the proportion of core and wall was 2:1, the concentration o f wall material was 0.5%, the proportion of wall material composition was 1:1, and pH was 6.0.Finally, the embedding rate of products was 78.6% and the yieldwas 89%, the density was 0.625g/cm3.
