Near-infrared fluorescent dyes have widely used as fluorescence probes, and it’s spectrum is located in near infrared region(600-1000nm) where a biologiacal matrix exhibits the least absorption and autofluorescence, weak background interference and strong SNR(signal to noise ratio). Heptamethine indole cyanine dye is a class representative of near-infrared fluorescent dye which consists of indole heterocyclic, heptamethine chain and the N-substituent side chains. It has extensively used in labeling protein, detecting intracellular metal cations, targeting tumor and other biological aspects.Lisosome(LP) has been widely applied as an effective drug carrier due to its good biocompatibility, biodegradability and subtle toxicity. liposome-encapsulated fluorescent dye can trace the biodistribution of the liposome and help evaluating the treating effects.With sulfonated phenyl hydrazine as raw materials,1,4-butane and benzyl bromide with different substituents as alkylating agent,2-chloro-l-methyl-3-(hydroxymethyl) cyclohexene as condensing agent, synthesized five heptamethine indole cyanine dyes, which al have preferable watersolubility and light stability, spectral range of them are between 780-800nm,and on the basis of the dyes synthesized one kind of dye with large stokes shift. A new near-infrared fluorescent liposome has been successfully designed and preparaed through loading a conjugate which contains the near-fluorescent dye having large stokes shift and 1,2-Distearoyl-Glycero-3-Phoshphoethanolamine(DSPE), laying a foundation for the further study.
