Based on Rana skin collagen peptide(LWT) as raw material and the method of combining the chemical modification(glycosylation graft) with the physical modification(ultrasound technology),the article doesn’t only modify the LWT molecules through the medium,response surface optimization,but also did some performance study and characterization analysis on LWT-CMCS at the same time. The results were as follows:1. Respectively disposed the LWT by the method of adsorption of diatomite,activated carbon filtration, metal catalyst treatment and Chitosan Graft Modification Method, the chitosan graft modification method has been regarded as the best treatment method.2. The performance study made it clear that the performance of the CM-CS, which was synthesized by titration with acetic acid, has improved. And the fact that carboxyl methyl group has been introduced in CS molecule was indicated by the characterization analysis.3. Did single factor experiment analysis according to the influence of the reaction temperature,reaction time, pH, protein concentration and protein/ carbohydrate ratio of the system on graft reaction, then initially identified the conditions of grafting reaction under hygrothermal conditions are as the following: the reaction temperature is between 60 ℃-80 ℃; time is between 4h-16h; pH about 7.5; protein concentration is 20 mg·mL-1; the ratio of protein and sugar is1:1. Reinforced the grafting reaction by ultrasonic radiation and determined the optimum conditions for grafting reaction under ultrasonic condition after the using of response surface optimization. The result shows that the DG value of grafting reaction reached to 55.12% and the comprehensive index up to 82.38, where the comprehensive index of samples has no bad smell and activity was not affected, when system temperature is 73 ℃,ultrasonic processing power is 263.40 W, ultrasonic time is35.28 min, protein concentration is 21.95 mg·mL-1.4. The relation between the DG value of grafting reaction and comprehensive index y value identified two linear positive correlation, namely the higher the DG value of grafting reaction, the more the graft y value, and the better the method to remove the effect of LWT’s bad smell.5. Study and analyze the solubility, thermal stability, moisture absorption and moisture retention and emulsifying properties of LWT-CMCS one by one to show that LWT-CMCS had good performance mentioned above.6. Test LWT and LWT-CMCS respectively by UV spectrophotometer, electrophoresis,high performance liquid chromatography, infrared spectroscopy, and scanning electron microscope, then it can be determined that the sugar molecules was covalently bound into the LWT after the qualitative analysis.
