Objective: To prepare metformin hydrochloride-loaded nanoparticles and investigate their physicochemical properties, and to study the cellular uptake and mitochondrial uptake of drug and nanoparticles.Methods: The metformin hydrochloride nanoparticle was prepared by an improved double emulsion method with glycerin mono stearate as carrier material and soybean lecithin and PVA as surfactant. The high performance liquid chromatography(HPLC) method was established and the morphology of nanoparticles was observed by transmission electron microscope(TEM). Mean particle size and zeta potential were estimated. Entrapment efficiency and loading efficiency were determined by HPLC. Meanwhile, the cytotoxicity of metformin was evaluated by MTT assay, the cellular uptake and mitochondrial uptake of drug and nanoparticles was evaluated by high performance liquid chromatography.Results: The nanoparticle solution of metformin was clear with blue opalescence. The morphology of the nanoparticle was almost spherical and no conglutination from the results of TEM. The mean particle size and zeta potential of Metformin hydrochloride nanoparticle were(102.3±4.16)nm and(-21.25±4.89)mV, respectively. Entrapment efficiency and loading efficiency were( 26.25±2.59) % and(1.74±0.16)%, respectively. The nanoparticle was proved prromote the cellular uptake and mitochondrial uptake by high performance liquid chromatography.Conclusion: The double emulsion solvent evaporation(w/o/w) method is simple and reliable, the process has a good reproductivity. The outcome of our research provides a feasible path to redevelop of metformin.
