1 To prepare good molecular imprinted polymers(MIP) in nature, every kind of element and condition among the process that MIPs were prepared were optimized, and then detected by spectrophotometer. In conclusion, the MIPs and NIPs were prepared by MG as the template, acetonitrile: toluene=3:1 as the solvent, MAA as the functional monomer, EGDMA as the cross-linker, and there ration is 1:4:20. In addition the 10% acetic acid-methanol acted as the eluent and worked in the apparatus. Finally all the upper terms were applied to prepare the MIPs and NIPs that were apparent in structures.2 The coating MIPs should be detected in terms of absorbance, specificity and so on, to acquire the qualified detecting plates. Then the MIPs and NIPs were proved that their adsorbing capacity can stay stably, MIPs’ capacity was 840.370μg/g, and NIPs’ 370.470μg/g. And the capacity of MG is 1.868 times than CV and 9.190 times than PA. In addition MIPs could keep adsorption stably in many conditions, such strong acid and base. In short preparing better MIP is critical to the detecting plates,so according to the upper data the MIPs prepared can be coated to detect.3. In order to make the detecting plates used in practice, every parameter of the plates must be measured, determined and optimized to meet the national standard. According to the method combining ELISA and CLA, MG-MIP-96-wells detecting plates’ standard curve was y=-29197.49x+1945998.45(R2 = 0.9416), linearity range was 30-66.64ng/ml, the LOD was 3.355ng/ml, the intra and inter-assay CV were 7.38%-18.07% and 12.45%-17.52%, the intra and inter-assay recovery were 103.41%-144.45% and 100.07%-143.20%, and the CV resulted by matrix effects was 103%—143%. The crossover rates were 27.97% and 30.97% with specifically detecting the MG′s analogue, PA and CV. The experiment results show that the new detecting method has the great potential to develop and the value to apply in the practice, and the detecting results actually were the total residual quality of MG, CV, PA, et al.
