In recent years, There is increasing adverse events caused by drug abuse in the society, which endanger the safety of patients seriously. However, most detection methods developed so far are high cost and low efficiency, making it difficult to detect special drugs immediately. Surface-enhanced Raman spectroscopy(SERS) technique have many advantages such as high sensitivity and low water-disturbance, etc., therefore it has been widely applied in various fields such as food, medicines and materials. In this paper, the best conditions of SERS detection for special drugs utilizing metal colloid as SERS substrate were explored, and the detection of three special drugs by SERS has been accomplished, which for the supervision of special drugs are of important significance.Firstly, gold and silver colloid have been prepared by chemical reduction method, and the properties of which are characterized by field emission scanning electron microscopy(SEM) and UV-Vis-NIR spectrophotometer. Gold and silver nanoparticles with uniform size and spherical shape are obtained. A simple and highly efficient SERS detection mode with sample drips loaded on substrate with highly hydrophobic and reflectivity has been established based on metal colloid as SERS substrate.Then, the optimized conditions of the detection for diazepam injection were investigated and the detection was conducted using gold and silver colloid as SERS substrate. The enhancement effects of the detection for diazepam injection under different aggregation agents were researched using gold and silver colloid, and the best substrate and aggregation agent under the optimum conditions were determined. Besides, the SERS spectra of diazepam injection were collected continuously during experiment process, the optimal detection time was discussed and excellent reproducibility of SERS detection was guaranteed. Under the optimum experimental conditions, diazepam injection with various concentrations were detected, and the limit of detection was obtained.Finally, a simple and rapid SERS method to detect pethidine hydrochloride injection and morphine hydrochloride injection was established based on silver colloid. The SERS enhancement effects of silver nanoparticle induced by different aggregation agents were researched, and the aggregation agents with the best result were determined, respectively. Meanwhile, the reproducibilities of the detections on two drugs under the optimum conditions were also characterised. The SERS spectra of two drugs under various concentration gradients have been researched, the detecting limit of which have been obtained respectively. In addition, This method is simple, quick and accurate, indicating tremendous applying potentials of SERS in the detection of special drugs.
