Scientific bus location design and reasonable route optimization both play an important role in increasing the logistics management efficiency and opera-tion expenditure control in the whole bus service system. Traditional re-searches optimize location design and route planning separately, which may cause some danger to ignore the close connection between the two important problems.In this paper, firstly we propose a novel information entropy-based fuzzy minimax clustering (FMM) algorithm with good adaptability of different da-taset, and experiment evaluations are conducted to check the reliability and effectiveness of it. FMM can well adjust the clustering results by information entropy and the prior distribution of the dataset deviation.Then we employ FMM to address bus location optimization problem and obtain the optimal location strategy. Moreover, a bus route optimization model is formulated and an enhanced ant colony optimization (ACO) algorithm is designed to solve it.Finally, we apply the proposed approaches for enterprise’s integrated shuttle bus location and route optimization problems. The intergraded ap-proach is evaluated by a practical example and the results show that the pro-posed optimization model is acceptable and efficient for maintaining a well balance among the total staff travel cost, enterprise’s operating cost and loca-tions construction cost.
