Cloning and characterization of two key genes involved in biosynthetic pathway of flavonoids from purple Solanum tuberosum are presented in this text, to obtain a length of 953bp flavanone core fragment-3-hydroxylase (F3H) cDNA. Phylogenetic tree analysis revealed that F3H differentiation earlier than ANS and FLS. The results show that the flavonoid biosynthetic pathway enzymes were grouped into different functions corresponding branch..The full-length cDNA sequence of CHS gene with 1370 bp was isolated from purple S. tuberosum (designated as StCHS) using RACE and PCR, which contained a 1170 bp ORF encoding a protein of 389 amino acids with a calculated molecular mass of 42.46 kDa and an isoelectric point of 6.72. The deduced StCHS protein showed high identities to other plant CHSs via multiple alignments. RT-PCR analysis revealed that StCHS expressed differentially in different tissues of S. tuberosum with the highest expreesion in stalks and leafstalks. The expression of StCHS was induced by gibberellic acid, but it had no significant difference among the experimental groups. The expression levels of StCHS was strongly induced by sucrose, among which the highest transcript level of StCHS was found when treated with 120 g/L of sucrose. |
