Salvia miltiorrhiza Bge. belong to Labiatae sage genus, cryptotanshinone was extract from Salvia miltiorrhiza Bge. and S. przewaquskiiMaxim root, which have the quinonoids compounds. The material was pick from shangluo shaanxi province(TSLY Plant Pharmaceutical Co,Ltd.). Applied HPLC measured the cryptotanshinone contents as the investigation index, by single factor experiment and orthogonal test, examines the influence factors of extraction efficiency and level, simultaneously the macroporous resin adsorptive and purification extract of cryptotanshinone was done. This experiment is designed to find a optimal cryptotanshinone extraction technology and using macroporous resin purification cryptotanshinone, and support the factory made a guidance in extraction and separation. The main conclusions as follows:1. Previous salvia miltiorrhiza processingThe raw material of salvia miltiorrhiza roots after different previous drying treatments,the results showed that: the properties of salvia miltiorrhiza roots have significant differences; Set the content of tanshinones as the key facter, target objects cryptotanshinone and root skin color is irrelevant; 60℃drying processes of salvia miltiorrhiza materials get the highest content of cryptotanshinone. This experiment is the best processing method.2. Extraction technology of Cryptotanshinone60℃drying processes for experimental materials of salvia miltiorrhiza materials. Then we adopt two kinds of commonly tanshinones extraction methods: ultrasonic extraction and ethanol refluxing extraction, and the results showed that the ultrasonic extraction is better. Reference other papers, compared the ultrasonic extraction and enzyme auxiliary ultrasonic extraction. Results showed that the experiment used enzymeis was not good as the desired effect, and also not fit the economic benefits, so we choose the method of ultrasonic extraction for the final way. The ultrasonic extraction have already determined, later we used the orthogonal way as well consider of four key factors optimize the ultrasonic extraction.Results: the solid-liquid ratio is 6, the ethanol concentration is 80%, the extraction time was 45mins, and the extraction times were 2 times.3.Separation and Purification of CryptotanshinoneTo optimize the optimum conditions for the isolation and purification of cryptotanshinone from tanshinone extract. The optimized conditions were the selected, which were LSD-40 type macroporous resin, pH value was 3, time of duration was 3h, 80% ethanol gradient elution, elution volume of 150mL. Can be increased from the concentration of tanshinone from the extract of about 21% to more than 45% under these conditions, the concentration increased about 2.14 times.
