To understand the expression of Sox9 and P450 aromatase(P450arom) in the chicken embryonic gonadal development, and determine whether they can be used as molecular markers of the early chicken embryonic gonadal development and to identify the gonad sex in sex reversal individuals, semi-quantitative RT-PCR and whole mount in situ hybridization (WISH) were used to detect the expression of Sox9 and P450arom in E6.5~E12.5 female and male chicken gonads, respectively. The results obtained in this study are as follows:1. Semi-quantitative RT-PCR was used to analyze the expression profile of Sox9 and P450arom in E6.5~E12.5 chicken embryo gonads, at the same time to analyze the expression difference of different sexs in the same period or different periods in the same sexs. The Sox9 gene expressed specifically in E6.5~12.5 male chicken embryonic gonads. P450arom was detected expression in E6.5~E12.5 female chicken embryonic gonads, while was not found in E6.5~E10.5 male embryo except E12.5 ones. The P450arom gene expressed specifically in E6.5~E10.5 female embryo.2. The spatial expression pattern was used to analyze Sox9 gene in the normal chicken embryonic gonads and treated (over-express Smarcel gene) ones, respectively, showed that the Sox9 gene mainly express in E6.5~E12.5 normal chicken male embryonic gonads, while not detected expression in female embryo. Besides in E6.5-E12.5 male chicken embryonic gonads, Sox9 was also detected expression in E8.5-E12.5 embryo.3. The spatial expression pattern was used to analyze P450arom gene in the normal chicken embryonic and treated (over-express Smarcel gene) ones, respectively, showed that the P450arom gene mainly expressed in E6.5~12.5 normal chicken female embryonic gonads, while not detected expression in male embryo. Besides in E6.5~12. 5 female chicken embryonic gonads, P450arom was also detected expression in E8.5~E12.5 male embryo.The results indicated from the two detection methods and the research of WISH to the treated chicken (over-express Smarcel gene):P450arom and Sox9 genes were detected expression in the male or female chicken embryonic gonads, respectively; Sox9 gene expressed specifically in E6.5-E12.5 chicken embryonic gonads, so it could be used as molecular markers in early male chick embryo gonadal development. P450arom gene expressed specifically in E6.5~E10.5 female chicken embryonic gonads, and also could be used as molecular markers in early female chicken embryonic gonadal development.
