MiR-483-3p was highly expressed in fetal liver, it was located in the intron of IGF2. MiR-483-3p can promote cell proliferation and inhibite cell apoptosis through targeting BBC3 and PUMA, in coordination with IGF2 in cancer, but the fuction of miR-483-3p played in growth was unknown, especially the role of miR-483-3p in regulation of fat deposition. The main experimental methods and results of this study are as follows:1. The expression patterns of miR-483-3p and IGF2 were characterized by real-time polymerase chain reaction in 10 tissue samples. miR-483-3p and IGF2 had the same expression trend, suggesting that they maybe co-regulated and the fuction of miR-483-3p and IGF2 were associated.2. We found a SNP in the primary transcript of miR-483-3p, and it was significant associated with immune and fat traits by PCR-RFLP, implying the function of miR-483-3p is diversed.3. PPARBP and CTNNB1 were found in the predictional target genes of miR-483-3p by the soft of hoctar, and miR-483-3p can specially bind the 3’UTR of PPARBP and CTNNB1, the mRNA and protein expression of CTNNB1 was down regulated in 3T3L1 after miR-483-3p transfection, while the protein expression of PPARBP was not changed.4. When the pigs were feeding with clenbuterol, the expression of miR-483-3p and CTNNB1 was upregulated in liver and muscle tissues, though there is no change in IGF2; miR-483-3p, CTNNB1 and IGF2 was upregulated in heart and adipose tissues.In short, miR-483-3p can target the gene of CTNNB1, it maybe play important roles in fatty deposition, and the expression of miR-483-3p can be regulated with and without the dependence of IGF2 in this process. | | Keywords/Search Tags: | pig, miR-483-3p, IGF2, CTNNB1, PPARBP, fat deposition, expression regulation | PDF Full Text Request | Related items |
