Echinococcus granulosus has extensive intermediate host in Qinghai province,due to the diversity host. Echinococcus granulosus could have specific mutation inthe long-time evolution process. Yak, as a unique species in Qingahi-Tibet plateau, isa unique intermediate host of Echnicococcus granulosus. Whether Echnicococcusgranulosus from yaks could cause new genetic type or local strains is unknown atpresent.To understand the situation and the prevalence strain of Echinococcusgranulosue of yaks in Qinghai province, eight counties around the province wereselected. Using2mitochondrial target genes(COIandNDI) to analyzing mitochondriagene polymorphism from this counties. As a result of the amplification of partialsequence of mtDNA, fragments of324bp (G1) and380bp (G6) from COI gene andfragments of428bp (G1) and462bp (G6) from NDI gene was obtained. All thenucleotide sequences were found no insert and delete mutation, only base transitionand transversion can be found and base transition was obviously more than basetransversion. The sequences of mtDNA COI gene show that there were7differentsequences existed and5mutations were found in this different sequence, the genetichomology of Echinococcus granulosue G1type isolated with the completemitochondrial G1genome referenced in GenBank(AF297617)were99.1%~100%,and there were no significant of the homology index from the sequence among thiscounties; And the mtDNA NDI gene show that there were8different sequencesexisted and7mutations were found in this different sequence, the genetic homologyof Echinococcus granulosue G1type isolated with the complete mitochondrial G1genome referenced in GenBank(AB208063)was99.2%~100%, and there were nosignificant of the homology index from the sequence among this counties.Furthermore, the genetic homology of Echinococcus granulosue G6type isolatedwith the complete mitochondrial G6genome referenced in GenBank(AF297617)was100%(COI gene)and99.6%(NDI gene).According to the COI gene sequences and NDI gene sequences, the phylogenetictrees were constructed, by comparing this trees, confirmed the reliability ofexperimental results. All, we have done in this experiment, are good for the classification ofEchinococcus granulosus of yaks and also can provide basis data for the preventionand control of echinococcosis in Qinghai province. |
