In the present study, a molecular method was established for accurate identification of batspecies, sequence analysis of cytochrome b gene has been used to identify13bat species withinfour distinct families, Hipposideridae, Rhinolophidae, Vespertilionidae and Pteropodidae.Phylogenetic analysis of those bat sequences was highly coincident with but more accurate thantraditional morphological method therefore this method can be used to differentiate the specieswith minor morphological differences. The new genetic identification method can be applied inbat ecology research.Bats are considered as important animal reservoirs of viruses including many pathogenic tohumans. Group A rotavirus (RVA), as a typical zoonotic virus, is the most important causativeagent of viral diarrhea in young children and animals. In a viral metagenomic analysis of batsfrom Yunnan province,26reads were noted to group A rotavirus (RVA). Further RT-PCRscreening on bat samples and in vitro viral isolation on cell cultures confirmed the presence of anovel RVA, named as RVA/Bat-tc/CHN/MYAS33/2013/G3P[10]. Full genomic sequenceanalyses showed that MYAS33possessed the genotype constellationG3-P[10]-I8-R3-C3-M3-A9-N3-T3-E3-H6. Multiple sequence alignment showed thar MYAS33shared the highest gene homology with bat rotavirus MSLH14(94.7%in VP3,93.7%in NSP1,NSP2(95.3%), NSP5(97.4%)), human rotavirus CMH079(VP4(94.8%),VP6(94.3%),NSP4(97.6%)), horse rotavirus E3198(VP7(92.9%),VP2(95.4%)), monkey rotavirus TUCH(VP1(92.9%), NSP3(91.4%)). However, MYAS33was completely distinctive from the first rportedbat rotavirus KE4582in genotype constellation, while there are ten segments(except VP4gene)were the same genotype with first isolate bat first rotavirus MSLH14. The typical rotavirusparticles can be seen under the electron microscope using virus cell culture. The11genesegments of these two isolated bat rotavirus has been completely separated by PAGEelectrophoresis, and observed that VP6, NSP3, NSP4segments size of these two strains aresignificant different. By using the virus to inoculate mice we obtained antiserum and it can beused to serological surveys. In this study, we isolate a bat rotavirus with a rare G3P[10] type,indicate the diversity of bat rotavirus and the multiplicity of this rare rotavirus. The presentstudy highlights the potential role of bats as reservoirs for RVAs.
