| BALOs(Bdellovibrio-and-like organisms) are a kind of bacteria which can lyse othergram negative microorganism and reproduce themselves by using the material of hostbacterium.It mainly preys on gram negative bacterium completely, especially the vibrio whichis the dominant factor causing aquatic disease.With its specialty,it is widely applied in theaquaculture and attract more and more researchers.For this thesis,we research the Bdellovibriobacteriovorus BDHM01by using DNB double layer agar phating technique,negative stainand Transmission Electron Microscope, acceleration experiment in different temperature,PCR-DGGE finger-print technique.The following are the main results:(1) The isolated Bdellovibrio bacteriovorus can form the plaques which is inerratic,simple andtransparent and shows pole or egg shape with a thin and long flagella.By singal factorexperiment and orthogonal test, it has been found that the optimal temperature, pH valueand salinity are respectly30℃、7.0、30‰, which are in coincidence with some knownBdellovibrio.(2) The second part shows that we conduct the acceleration experiment in differenttemperature like25℃、37℃、45℃、55℃、65℃in order to identify the shelf life ofBDHM01micriobial preparation with the help of Arrhenius formula by mixing BDHM01with different protectants like glutamate, glutamate&sucrose, raffinose, glucose, glycerol.The results read that mixed protectants of glutamate and sucrose can effectively extend theshelf life of BDHM01microbial preparation and the concentration of BDHM01can stillmaintain108after8weeks experiemts.(3) The third part we do the research that what effects the application of BDHM01microbialpreparation have on the structure of ablone gut microflora.The results shows that theexperimental group have positive impact containing change of dominant bacterium afterapplication of BDHM01microbial preparation,while the control group have negativeeffect which means the dominant bacterium almost don’t changed. |
