Physcomitrella patens is new model plant of study on genesfunction about plants. The study on the interactions betweenplants and pathogeny microorganism, with Physcomitrella patensas the material,can take advantage of the benefits ofPhyscomitrella patens for the study of molecular biology tofigure out function mechanism in plant disease resistancereaction and also to will broaden the knowledge of theevolutionary process of early land plants in plant diseaseresistant mechanism. The research begining with cellmorphology can lay foundation for the study of interactionsbetween Physcomitrella patens and pathogenic fungus on themolecular level.This experiment observed the interaction of Physcomtrellapatens with Botrytis cinerea on the morphological change aboutthe appearance of Physcomitrella patens, cellular level andsubcellular level,so we can master infection process ofBotrytis cinerea roundly,quantize the infection process,andestablishevaluation criterion about suffering injury levelof bryophyta.It will provide the entry points for the reachabout defense response of Physcomtrella patens on molecularlevel further. Physcomtrella patens to get vaccinated with Botrytiscinerea after1day,there are some brown spots on the leavesof Physcomtrella patens,and after11days,most plant die.Theresults of light microscope observation showed that,aftervaccinated1day, the spores of Botrytis cinerea germinated andformed appressorium. after vaccinated2days,hypha started tointrude into the cell’s internal and intercellular space.At thelater period of Botrytis cinerea infection, there wascollapse in some leaf blade cells.After observation In the transmission electronmicroscope,we found there wereinvasion of bolts at the bottonof appressorium,some intruded into the cells of the plantsdirectly;the others formed inflate hypha in the cell wall andextended in the cell wall, then intruded into the the cell’sinternal.The result aslo showed that the hypha in the cellscould infect the neighbouring cell.In the preliminary stageof infection the plasmalemma of infected cell shrinked,andat later period, cytomembrane and most organelledisintegrated.Furthermore,we can found that plant cell wallwas thickening next to the invasion of bolt, and some papillaindeed.The morphologic change of Physcomtrella patens is verysimilar with defense response of vascular plants,so the thickening of the cell wall and formation of papilla both areconservative defense response mechanism of plant.After KMnO4staining, there is absence of lignin in infectedbryophytes,this also verified they are conservative defenseresponse mechanism of plant.After detection of H2O2, abundantH2O2were found in the infected cells. H2O2maybe take part information of lignin to reinforce the cell wall and resistinfection of pathogenic bacteria. This kind of defense responsewas also observed in nonvascular plants. So thephysio-biochemical changes as mechanism of resistance mayexist in the early land plants.
