Newcastle disease (ND) is caused by Newcastle disease virus which can bringa high mortality rate, acute and contagious disease. Newcastle disease (NDV) hascaused significant loss to the aquaculture industry. NDV was first isolated and namedby Doyle in1927. The virus can spread among different species, it can spread in thesame species, also can be in two different species. it is because ofthis transmission, the virulence of the virus is constantly changing, so there havebeen some new strains of different virulence appeared. Newcastle disease virus hostrange is wide, almost all domesticated and wild birds can be infected with the virus, isalso reported to have human and rodent animal and mammals can also be infected, butchicken, turkey, pheasant, dove, geese, birds also can infect, waterfowl has someresistance to Newcastle disease virus.The research object of this experiment is a strain of Newcastle diseasevirus from pigeon. This experiment mainly analyzes the virus F gene type, and judgethe genotype and genetic relationship.In order to obtain this, isolate and identify thePigeon Newcastle disease virus,design the RT-PCR primer, RT-PCR, cut the glue ofthe extraction PCR product, connection and transformation, plasmid extraction, sendthe plasmid to Shanghai biotechnology company for sequencing, the sequencingresults compare with the nine different gene sequences published by the NCBI anddraw the genetic phylogenetic tree,these are necessary.Then according tothe published Gene Bank NDV all gene sequence,15pairs of RT-PCR primerswere designed.following the above steps, finally sequencing, the sequencing resultscompare with some different NDV all genes sequences which have been publishedin the NCBI, drawing the phylogenetic tree, determine its molecular biologicalcharacteristics. After the analysis of sequence results, the Newcastle diseasevirus strains genotype from pigeon can be determined that the strain belongs to theweak strain.The experimental animals are chickens and pigeons. The Newcastledisease virus from the pigeon was injected to the chickens and pigeons. The incidencewas observed, and the pathological changes of tissues and organs were made frozensections.Then did HE staining and IFA.Observed in the results under the microscope,it can determine the distribution of the pigeon Newcastle disease virus.
