To explore the adaptive mechanism of mitochondria of Yak brain in hypoxiaenvironment, and provide basic research material for hypoxia adaptationmechanism research of Yak. We mainly use Gene cloning techniques. Cloning andsequencing studies was given to the coding sequence of the CDS sequence ofadenine nucleotide translocator(ANT) in mitochondria of brain of Qinghai DatongYak using Real Time PCR, the relative expression of ANT1and ANT2had beenmeasured in Yak brain compared with plain cattle by SYBR Green Real Time PCR.The result:1.We measured the CDS sequences of ANT1and ANT2gene. Thesoftware DNAman analysis result showed that Yak and cattle had similar CDSsequences of ANT1or ANT2in addition to several bases. ANT, as a member ofmitochondrial solute carrier family, is highly conservative.2.We measured therelative expression of adult yak ANT1and ANT2gene in different region of brain.The results showed:①Both express ANT1and ANT2,the expression of ANT1and ANT2in different areas of yak brain were higher compared with cattle, thedifference was statistically significant(P<0.05);②The expression of ANT1andANT2gene in the same region of the brain of yak were higher compared with cattle,the difference was statistically significant(P<0.05);③in the same area of the brain,the expression of ANT1in yak and cattle were higher compared with ANT2, thedifference was statistically significant(P <0.05).Conclusions:1. the CDS sequence of yak ANT gene is highly conserved andthe locus mutation of gene is not obvious.2. the expression of ANT1and ANT2gene in yak was higher compared with cattle, indicating that ANT plays a role inthe adaptation to hypoxia.3the expression of ANT1in yak and cattle were highercompared with ANT2. The result showed that ANT1genes may be more conduciveto yak mitochondrial adaptation to hypoxia.
