This study aims to investigate the expression levels of heme oxygenase HO-1andBLVRA in the interstitial part of uterine tube and compare the different expressionlevels in Lushi chickens and Gushi chickens, thus discovering the formationmechanism of green shell eggs.To conduct the experiment,5Lushi chickens laying green shell eggs and5gushichickens laying brown eggs, all of which were24-week aged, were randomly selectedand the uterus of each chicken was collected. Then the total RNA was extractedaccording to the method provided by the TRial RNA kit, which was reverselytranscribed into cDNA later. After that the upstream and downstream primers weredesigned, and the pigment-associated genes, HO-1and BLVRA were chosen to be thetarget genes, and GAPDH the reference genes. Finally,the transcription levels of HO-1and BLVRA in different breeds of chickens were analyzed using real-time fluorescentquantitation PCR.Results showed that after analyzing the extracted total RNA, it was found that thebands of28S and18S were relatively brighter, and the band of5S was not clear, andthis meant that extracted total DNA was of good quality. The real-time fluorescentquantitation PCR indicated that the expression levels of HO-1in Lushi chickens andGushi chickens were0.1581±0.0278and0.0299±0.0050respectively, and those ofBLVRA were0.0392±0.0971and0.6315±0.0059respectively.In summary, it could be concluded that with the application of independentsamples T test, the HO-1mRNA expression level in the uterus of Lushi chickens ishigher than that of Gushi chickens (P<0.05), and that the BLVRA mRNA expressionlevel in the uterus of Gushi chickens is significantly higher than that of Lushi chickens(P<0.01). Therefore, the expression levels of H0-1and BLVRA are in close associationwith the formation of green shell eggs, which could provide theoretical reference forfuture research.
