Fusarium wilt of banana is a devastating vascular systemic fungal disease. Its pathogen is Fusarium oxysporum f. sp. cubense that spread by soil, irrigation water, rainwater and infected banana materials. The pathogen is a kind of typical soil inhabitant and it can survive in the soil for a long time which is difficult to be effectively controlled. The fusarium wilt of banana had frequently happened in banana production place of the word recently. At present, it only had a little of research about banana and Fusarium oxysporum interaction of molecular mechanism. But the molecular mechanism of banana and Fusarium oxysporum interaction research have very important value and significance for controlling Fusarium Wilt of banana. We use high-throughput Digital Gene Expression profile sequencing of technology in this study for researching the differential expressed gene and investigating the molecular mechanism of banana and Fusarium oxysporum interaction. The results were as following:1) The digital gene expression sequencing got24,914,334useful reads. The total reads number about the four stages of0h、4h、24h、6d were5,888,335,5,893,272、6,964,634、6,168,093. Among those datum, the reads can be mapped to reference gene and reference genome respectively were3,495,218、3,283,752、4,213,136、3,635,245and4,686,660、4,704,901、5,602,183、4,910,086. And the unique mapped reads were3,432,075、3,222,116、4,126,345、3,570,754and4,475,367、4,347,173、5,405,161、4,565,327.2) We acquired Oh (BCK)、4h(B4h)、24h(B24h)、6d (B6d) of the DGE by the digital gene expression sequencing. we had screened differentially expressed genes by the differential expression ratio larger than2. The results show that:BCK VS B4h, there were4072genes of up-regulation and2369genes of down-regulation and with the ratio to the total number of differentially expressed genes as63.22%and36.78%when banana leaves were inoculated with Fusarium oxysporum after4hours; BCK VS B24h, there were2355genes of up-regulation and1267genes of down-regulation and with the ratio to the total number of differentially expressed genes as65.02%and34.98%when banana leaves were inoculated with Fusarium oxysporum after24hours; BCK VS B6d, there were4899genes of up-regulation and2289genes of down-regulation and with the ratio to the total number of differentially expressed genes as68.02%and31.98%when banana leaves were inoculated with Fusarium oxysporum after6days. We had screened differentially expressed genes by the differential expression ratio larger than4.The results show that: BCK VS B4h, there were1721genes of up-regulation and1288genes of down-regulation and with the ratio to the total number of differentially expressed genes as57.19%and 42.81%when banana leaves were inoculated with Fusarium oxysporum after4hours; BCK VS B24h, there were690genes of up-regulation and427genes of down-regulation and with the ratio to the total number of differentially expressed genes as61.77%and38.23%when banana leaves were inoculated with Fusarium oxysporum after24hours; BCK VS B6d, there were1835genes of up-regulation and1383genes of down-regulation and with the ratio to the total number of differentially expressed genes as57.02%and42.98%when banana leaves were inoculated with Fusarium oxysporum after6days.3) We have screened185significantly differentially expressed genes by analysing plant and pathogen interaction pathway of BCK VS B4h、BCK VS B24h、BCK VS B6d.4) We selected20significantly differentially expressed genes that were verified by semiquantitative RT-PCR for determining the authenticity of sequencing results of DGE. The results of this study will help to reveal the molecular mechanism of banana infected by F. Oxysporum, because these genes probably were concerned with the course of the interaction.
