Chitinase is an important hydrolyzase, and it can catalyses the reaction of hydrolyzing the chitin into N-acetylglucosamin (GlcNac) and (GlcNac)n. Chitinase universally exist in plant and microorganism. The cell wall of fungi have chitin. Chintinase is thought to be an enzyme related to fungi disease resistance, it can degrade the cell wall of pathogenic fungal hosts, suppress mycelium growth, spore germination and germ tube elongation. So, chitinase play a role in plant defense against pathogenic fungal hosts.Thermomyces lanuginosus are widely distributed soil-inhibiting fungi of considerable interest producers of thermostable enzymes, including chitinase, endocellulase, glucoamylase, xylanase, cellulase, and laccase. The fungi have great value in research and application.In my lab, a chitinase gene Lan425was isolated from Thermomyces lanuginosus. The full length of Lan425cDNA gene is1454bp, which contained an ORF of1278bp encoding425amino acids.To investigate the disease-resistance of a chitinase gene Lan425from Thermomyces lanuginosus in plant, the chitinase gene Lan425were subcloned into the expression vector pCAMBIA1300downstream of the35S-CaMV promoter. And then the constructs were introduced into rice via Agrobacterium tumefaciens EHA105and verified by PCR, RT-PCR. The results indicated that transgenic rice plants could enhance disease-resistance than the wild plants at the seedlings growth stage. The chitinase-transformed plants exhibited increased resistance to stalk infection with the fungi Thanatiphorusu cumeris.We have gotten seeds of To transgenic rice and rice seeding of T1generation.
