To provide basic date for exploring the high flavonoid apple germplasm resources,the conservation and sustainable use of Malus sieversii f. neidzwetzkyana. resources and the creation of functional apple breeding theory and technology system, the content of flavonoid and gene expression intensity related to flavonoid biosynthesis during the fruit growth, comparative analysis of single fruit weight, hard crispness, soluble solids, total phenolic and anthocyanin content, antioxidant ability, the content and components of flavonoids of the "Hongcuil","Hongcui2","Hongcui4","Hongcui8","Hongcui10","Baicuil"and"Zihong2" functional apple fruit from F1progenies of the cross of’Fuji’and Malus sieversie. were detected and compared with’dehongcui’which was from frogein and’Golden Delicious’,The main results indicated that:1) The changes of flavonoid content between the functional apple and "Golden Delicious"had declining trend during the growth period. The flavonoid content in4functional apple was significantly higher than control "Golden Delicious",among them,125days after flowering date," Zihong2" and" Hongcui1" had the highest flavonoid content3.13mg/g and3.00mg/g,which was4.08and3.9times of0.77mg/g in "Golden Delicious".2) The expression intensity of structural genes PAL etc related to flavonoid synthesis in4functional apple is generally higher than control "Golden Delicious "during the fruit growth period,and there is obvious difference between4functional apple, among them,in the middle and later growth period,the expression intensity of PAL,4CL,CHS,CHI,F3H,DFR and LDOX in "Hongcui1"is higher than others;in the later growth period, the expression intensity of FLS is higher in "Hongcui1",and "Zihong2" has a increase trend of the higher expression intensity of4CLand LDOX.3) The expression intensity of transcription factor between materials was significantly different,among them, the MYB1had the highest expression intensity at95days after flowing date,showed a rapid downward trend in "Hongcui1";At late growth period of "Zihong2",The expression intensity of MYB1and BHLH33showed a increase trend,and had maximum at125days after flowering date.4) The fruit size, fruit shape index, hard crispness and soluble solids content had no correlation with red coloration degree, and there was no obvious regularity among the differences of material; The red strains "Hongcuil" and "Hongcui10",which of the red area was nearly1/3-1/2, and their flavonol、anthocyaninn、total phenolid conten and antioxidant capacity were extremely significantly higher than those of reddish apple "Hongcui8""Baicuil" and the control. The flavonoid content of "Hongcuil" and" Hongcui10" was4.4and4.32mg/g, which was respectively5.1,2.8,2.4and1.6times of the"Dehongcui"," Golden Delicious","Baicuil" and "Hongcui8".5)20flavonoid constituents including Cy3-0-galactoside, were detected from the6apple varieties (lines) fruit. The red strains "Hongcuil" and" Hongcui10"had all of the15flavonoid constituents, extremely significantly higher than10flavonoid constituents of reddish apple"Hongcui8" and "Baicuil", and8flavonoid constituents of the control.
