| Nonylphenol (NP) is a nonionic surfactant, which brings convenience to people,simultaneously brought many negative ecological effects, such as estrogenic effects.In our present study, we selected the prawn, M.rosenbergii, as the experimentalmaterial. We studied the pharmacokinetics of NP in the tissues, effects of NP onorganization structure and effects of NP on the expresstion of gonad-related genes ofM.rosenbergii by bath. The main results are as follows:1. The pharmacokinetic of NP in the tissues of Macrobrachium rosenbergiiThe tested prawns were exposure to1mg/L concentration of NP solution with3days. The tissues were sampled at0,0.5,1,3,6,12,24,48,96,192h after bathtreatment and the concentration of NP indifferent tissues samples were measured byLC-MS method. The pharmacokinetic parameters were calculated according tonon-compartment model using WinnolinV6.0special software. The concentration-time curve of NP appeardouble peak phenomena. The time to peak concentration(Tmax) of hepatopancreas, ovary, eyestalk and muscle are3h,3h,3h and1h. Thecorresponding peak concentration(Cmax) are13447.95,4263.66,2860.42and1299.06μg/kg respectively. This showed that NP was absorbed anddistributed rapidly inM.rosenbergii. The area under the curve (AUC) are512975,240594,44878and11700(μg/L h) and it revealed that the tissue with highest absorptive capacity of NPwas hepatopancreas, followed by ovary, eyestalk and muscle. The mean residencetime (MRT) are40.65,54.82,61.53and32.97h and it revealed that NPmolecule remains in eyestalk longest, followed by ovary, hepatopancreas and muscle.The elimination half-life (t1/2) were28.5,33.2,85.0and38.6h respectively. Theresults shows that NP was eliminated faster in hepatopancreas and ovary, buteliminated slower in muscle and eyestalk. 2. Effects of NP on histological changes of Macrobrachium rosenbergiiThe tested prawns were exposure todifferent concentration of NP solution with20days. Meanwhile, the effect of NP on histological changes of testis, ovary andhepatopancreas were studied by paraffin sections method. The results showed that thehistologic structure of lowdensity group (1μg/L) have no significantdifference withcontrols. Medium (10μg/L) and high concentration (100μg/L) NP groups showedthedegree of Microstructuredamage aggravated with the increase of exposuredose andexposure time. NP could promote ovarydevelopment, the velocity ofthe ovarydevelopment was accelerated and the ovary reproductive cycle was shortenas the the increase of exposuredose and exposure time.3. Effects of NP on gonad-related genes (Mar-SGP-B, VTG, VgR) expression ofMacrobrachium rosenbergiiThere was no significantdifference between the lowdensity group (1μg/L)and control group. Medium (10μg/L) and high concentration (100μg/L) NP groupshad a inhibitory effect on the expression of Mar-SGP-B and a increased effect on theexpression of VTG and VgR. Overall, the high concentration NP groups was muchmore effective than the medium concentration NP groups. There is a significantdose-response and time-response relationships between NP and the expression ofMar-SGP-B, VTG and VgR. |
