Transfer factor (TF) is a low-molecular-weight lymphocyte extract capable oftransferring antigen-specific cell-mediated immunity to T lymphocytes. It is a novel immuneactivator and has function of enhance immune. In currently, it has been used successfully asan adjuvant or primary therapy for immunodeficiencies, allergies and autoummune diseasesof poultry. However, it is a confusing to understand the composition of transfer factor.In this reaserch, freeze thawing method was used in making transfer factor oral solutionfrom chicken spleen. And then, the peptide content and molecular weight, composition andreceptor activity were determined by Folin phenol method, SDS-PAGE electrophoresis,matrix-assisted laser desorption ionization time of flight mass spectrometry(MALDI-TOF-MS), liquid chromatography-mass spectrometry (LC-MS) and E rosettemethod, respectively. Meanwhile, it’s application of the clinical effect was evaluated.As a result, the preparation of chicken spleen transfer factor oral solution polypeptidewas1570.8μg/mL, the molecular weight polypeptide of the chicken spleen transfer factororal solution was9213Da, the activity of chicken spleen transfer factor was10.92%, themolecular weight polypeptide was9213Da and the molecular weight of the peptide LC-MSwere ranged around800-7476Da. There were581kinds of polypeptide species, including Tinhibitor of apoptosis related protein (TIAL1), Thymosin β4(TMSB4X), bactericidal/permeability-increasing protein (BPIL3), actin, hemoglobin and other biologically activeprotein. The results showed that of the transfer factor composition from the chicken spleen, ithas a beneficial to discover more active ingredients and mechanism in veterinary.In addition, the clinical trial to treat28weeks age laying ducks flavivirus infected duckuse Chinese medicine alone and combined with peptides (Baiduning) were administered. Theresult showed that the “Baiduning” combined with Chinese medicine has more effect inlaying rate evaluated for33days. And the average laying rates increased to a maximum of97.1%from60%,which significantly better than the Chinese medicine alone groups of90.8%、91.2%and untreated control of72.4%. It suggested that “Baiduning” combined withantiviral Chinese medicine can be used to treat laying duck flavivirus disease in28weeks ofage safely and efficiently. It has provided an exlperimental data for chicken spleen transferfactor as an oral medicine used in the clinical treatment of flavivirus infections.
