Acrosin is a multifunctional protein, plays a vital role in the breeding process. The main role of the acrosin inhibitor is to prevent premature release of sperm acrosin, damage sperm and take action to protect the epithelial cells of sperm and sperm transport in the female reproductive tract. However, the necessary condition for successful fertilization is active acrosome enzyme release, this process requires the ubiquitin-proteasome degradation pathway acrosin inhibitors, thus acrosome enzyme is released. Ubiquitin-proteasome pathway is an important intracellular protein degradation regulation system is in the cytoplasm and the nucleus is dependent on ATP conducted. First, the process is a target protein covalently coupled to a plurality of ubiquitin molecules to form the target protein polyubiquitin chains, that the target protein ubiquitin, and then is transported to the26s proteasome digestion degradation.To explore after the pig sperm capacitation acrosin inhibitors and the impact on the expression of the acrosin inhibition ubiquitination extent, whether the cause cryogenic ubiquitin-proteasome pathway is blocked, and cold shock to the acrosin inhibitors with to which the damage. The results showed that:the fresh sperm treatment group is higher than capacitation, acrosome reaction the expression of the acrosin inhibitor; capacitation and acrosome reaction treatment group the acrosin inhibition ubiquitination extent and PSMD4subunit expression were higher than fresh sperm group; however the cold shock treatment group appear unusual expression of acrosome enzyme inhibitors, the acrosin inhibition ubiquitination extent and PSMD4acrosome subunit expression levels decline.Conclusions are as follows:1. acrosome inhibitors occurs ubiquitination after capacitation or acrosome reaction.2. after capacitation or acrosome reaction, acrosome enzyme inhibitor combined with four ubiquitin molecules were completely degraded ubiquitin-proteasome system, the release of free ubiquitin molecules.3.19s subunit PSMD4accompanied capacitation, acrosome reaction handle its expression level gradually increased, after capacitation26s proteasome is activated, it changes the expression of the enzyme inhibitor acrosome opposite trend.4. inhibitors sperm acrosome cold shock occurred after abnormal expression and its extent and PSMD4ubiquitin subunit expression under the severe amount, probably because of the low temperature forecast brought acrosome inhibitor protein structure change or destruction, degradation of ubiquitinated proteins or change, PSMD4subunit damaged, so that the ubiquitin-proteasome pathway interference.
