Soaking Mengnong sainfoin pods in EMS (ethyl methane sulfonate) with different concentrations (0.0%,0.3%,0.7%,1.1%,1.5%) and different time (7h,15h,24h). Analysis the morphological characteristics, photosynthetic characteristics and main agronomic traits of the variant plants under the LD50dose, and combined with ISSR molecular markers technology, the paper explores the EMS mutagenic effects of Mengnong sainfoin, that will provide valuable basic materials for mutant library construction and high yield, high quality and hardiness sainfoin new cultivation system. The main findings are as follows:1. EMS delay Mengnong sainfoin emergence, when the comprehensive function of time and concentration gets22.5or more, the emergence and seedling growth was inhibited significantly by EMS(P<0.05).1.5%EMS soaking15h and1.1%EMS soaking24h, ie, treatment intensity was22.5to26.4, can be the appropriate concentration and time to establish the mutant library of Mengnong sainfoin induced by EMS.2. Through identify the phenotypic traits of the M1generation plants. There ar e15kinds of phenotypic variation types, including high plants, dwarf plants, lobula r denseness, lobular sparse, lobular big, lobular small, lobularl dark, lobular pale, re cumbent plant type, chlorophyll distribution uneven, lobular surface coat on, many branched inflorescence, flowers large, inflorescence of abnormal, basal leaves yellow ing. The main morphological indicators have significant difference(P<0.05), and the mutation rate was15.45%, it provides the abundant basis materials for the mutant1ibrary built of the M2generation.3. Observe and evaluate the traits of the morphological characteristics, photosyn thetic characteristics and main agronomic. The photosynthetic performance of lobular1dark, high plants, lobular denseness and flowers large are strong, and the main ag ronomic traits of lobularl dark, high plants and lobular small are outstanding. Amon g them, the high plants and lobular denseness are excellent at the photosynthetic pe rformance and main agronomic traits. They can be the superior materials of high yi eld, high quality and hardiness.4. Based on the10primers of ISSR markers, the polymorphism ratio of M1and M2generation respectively are88.89%,83.52%. In the threshold of0.659, M1generations can be divided into five categories, M2generation are divided into only three categories. That suggests that the genetic variation of M1generation is more rich, however, it can’t be all pass it on to M2generation.5. Compared with the contrast, the variation plants of M1and M2generation both have different loci, It implies that the phenotypic variation plants have taken place in a certain degree of genetic variation at the molecular level. Lobular pale has the most different loci and the largest degree of variation in M1generation; Lobular surface coat on has the most different loci and the largest degree of variation in M2generation.
