Illegal adding of salbutamol is a serious threat to food safety and bringsrisk to human. It also brings bad effects to both of livestock products breedingindustry and meat products processing industry. It is important to understand theeffect and mechanism of action of salbutamol, which would provide a scientific basisfor fast and accurate detection of salbutamol residues in meet. Because of theadvantages in genetics and biochemistry, especially many crucial genes with highhomology to higher organisms including human, the fruit fly is an important modelorganism for scientific research. In this study, we used fruit fly as the biologicalmodel to study the effects of salbutamol and their molecular mechanisms. This is thefirst time to study the effect of salbutamol on fruit flies. In this case, we studied thegrowth, behavior, life-span, anti-oxidative ability, DNA damage and the expressionlevels of certain genes of fruit flies fed with salbutamol. The results would providevaluable basis to develop the new detection methods of salbutamol.The basic approach is collecting and cultivating fly eggs with culture mediumcontaining salbutamol and evaluate their pupation rate and eclosion rate. The resultsshowed that under the treatment of salbutamol, the pupation rate and eclosion ratereduced with the increasing salbutamol, but there is no significant influence on theirdevelopment duration. The movement ability and the courtship behavior are adjustedby a series of complex mechanism. Based on the research of the mutant Drosophila,we studied the effect of salbutamol to the organism as a β-agonist. Base on theexperimental data, the mutant flies have become stronger and more active. Themating latency and mating duration are extended,but still completed within15to20minutes.After the cultivation of flies with salbutamol, the result of the measurement of theSOD, CAT activity and MDA indicated that with the concentration of salbutamolincreased, the content of MDA is significant increased (P<0.05); the activity of CATwas inhibited significantly (P<0.05); the antioxidant system was prompted and the activity of SOD increased significantly (P<0.05). May be the antioxidant system, acertain concentration of salbutamol could increase the lifespan of fruit flies.We cultivate flies with salbutamol and detect their DNA damage, cell divisionsand apoptosis. The DNA damage can be observed with the SCGE test which means itcould directly be triggered by salbutamol, and more serious of DNA damage werefound with the concentration of salbutamol increased. When the fly strains werecultivated in food with salbutamol, both the long-term and instantaneous treatmentshowed different levels of DNA damage, and which may due to their DNA damagerepair mechanism. The actin protein Actin-87E is activated by salbutamol, and therelated genes need further research.
