NAC gene family is an important specific transcriptional factors in higher plant, whichare involved extensively in plant growth, development and stress responding signal pathways.In this article, NAC transcription factors of Vitis vinifera were studied. By constructingthe phylogenetic tree, grape NAC protein sequences and some NAC protein sequences of otherplants were analyzed. Bioinformatics method and Real-time PCR were employed to study thecharacteristics of structure and biological function of NAC transcription factors. The mainexperimental results are as follows:1. Seventy one grape NAC protein sequences and Twenty two NAC protein sequences ofother plants were aligned and phylogenetic tree was constructed. We analyzed. The resultsshowed that71NAC family of grapevine were divided into five groups(ⅠⅤ). The groupⅠ included17grape NAC genes, concluded that the NAC transcription factors related to thegrowth of grape. All of the abiotic stress-related12NAC genes was fallen into the group Ⅲ,concluded Implied that those transcription factors might be involved in regulation of abioticstress.2. Twelve abiotic stress-related NAC genes was further analyzed for predicting geneticstructure, chromosomal location and cis-acting element in the promoter region.3. Twelve abiotic stress-related NAC protein amino acid sequence were analyzed onphysical and chemical properties, signal peptide, hydrophilicity/hydrophobicity.Phosphorylation sites, the secondary structure, conserved domain, motif and tertiary structure.The results showed that there are some relationship among the grape NAC transcriptionfactors related to abiotic stresses, and there are also many similarities among them on thestructure and functions. The NAC transcription factor of grape showed the characteristic ofhighly conservative N-terminal and diverted C-terminal in a NAC transcription regions.VvNAC59had a longer amino acid sequence than others. Its C-terminal showed a differentRicinBlectin2conserved domain from other NAC protein,and yet the similarity was nothigh. The biological function need to be studied further. The results of the analysis providedtheoretical basis and references for the further study on the growth of grape and the regulationand mechanism of NAC transcription factors.4. UniGene expression map of twelve grape NAC gene analysis showed that the EST ofVvNAC17was mainly expressed in leaf. Under exogenous hormones treatment and abiotic stress, The expression of VvNAC17was analyzed,Under treatments of different ET, ABAconcentration and damage stress, The expressions of VvNAC17varied increasingly The resultimplied that VvNAC17transcription factor might be involved in regulation mechanism ofplant stress responseThe reported results will provide references for further study of transcription factorregulation mechanism of plant growth, development and signal pathway.
