| The research is mainly breeding5R,6R small fragments translocation lines thatderived from the hybrids of wheat-rye5R/5A×6R/6A substitution lines and manifestedthe merit of rye, such as a large spike, disease-resistant and other good traits, itprovides the basis material for breeding new wheat that has the character ofhigh-yielding and disease-resistant.In this study, twenty lines of hybrids between wheat-rye substitution lines5R/5Aand6R/6A,10-1,10-2,10-3,10-4,10-5,10-6,10-7,10-8,10-9,10-10,10-11,10-12,10-13,10-14,10-15,10-16,10-17,10-18,10-19and10-20was identified bymorphology, cytology, molecular markers and GISH. The results showed that thechromosomes number of20lines were2n=42, and chromosome configurations ofmeiosis MI were normal in pollen mother cell; Six lines detected by rye-specificprimers PSC119.1were all contained genetic element of rye chromosome, six pairs ofspecific primers of SSR SCM138, SCM120, SCM268, CGG18, SCM180and CGG19could amplify5R or6R specific fragments of different sizes in twenty lines,respectively; The results of GISH showed that yellow or yellow-green signals of ryewere appeared in both ends of chromosomes of10-1,10-2,10-3,10-4,10-5,10-6,10-7,10-9,10-12and10-20. Above of results meant twenty materials were all wheat-ryetranslocation lines, and had the good traits of rye with morphology test, such as largespike, many spikelets, disease resistance and so on, so the six lines can be used as basicmaterial for wheat breeding, and had the important utility value for geneticimprovement and resistance research in wheat. |
