The Research of resin traits had become a hot issue due to the market in short supply. Forunderstanding of the resin traits on basis of molecular genetic, the published about cDNAof terpene synthases of pinus and the genomic of Pinus taeda was used in homologycloning of PeTPS-(+)Apin and PeTPS-(-)Apin of Pinus elliottii in this paper. The sequencehomology was analyzed, the structure and function was forecasted, they were used as thecandidate genes for associate with resin yield traits. Through the sequencing of DNAsequences about the24samples, we have found the SNP sites, gained the information ofnucleotide diversity, neutral evolution and LD recession levels, and also have filtered theSNP sites. In this research, we have measured the annual yield resin and the growth traitsof111families, and identified the related functional SNPs or haplotypes with thatphenotypic traits includes basic heritability except growth factors (W0), potentialheritability include growth factors (Wp) and the realize total yield (W). The main results areas follows:(1) The clonal TPS of Pinus elliottii was labile, with Molecular weight71.77-78.46ku,phosphorylation site37-39, locating in the chloroplasts. There are2-7irregular crispationand24-25α-helix on Biomolecular structure, and a cd00684conserved domain on tertiarystructure. It was surmised that a RR conservative domains existing in65-69a.a. couldbreak in N-terminal of the second R and combine with substrates,thus participated in theisomerization. The DDxxD conservative domains appearing in377-384a.a. which couldnot found a functional site. Two phosphorylation site were predicted at287-302and579-623a.a..(2) The gene PeTPS-(+)Apin was3175bp in length, the diversity of nucleic acids (π)and (θw) were0.00380and0.00354, which remains lower. The nucleotide sequence have astrong conservatism. The gene was experiencing the action of balancing selection, followthe neutral model in evolution. The level of LD was low, the value r2down below0.2atabout100bp, so a greater density marker has been need to. Only sites GT1131and CT2096have a weak association with the traits of the basic yeild (P≤0.1), with contribution value:0.15%and0.068%. Gene can explain0.218%phenotypic variation, but there may be afalse association, and we thought that there is only little relation between the gene and theyield traits.(3) The gene PeTPS-(-)Apin was4628bp in length, the diversity of nucleic acids (π)and (θw) were0.00277and0.00260, they were at low level. The nucleotide sequence havea strong conservatism. The gene was experiencing the action of balancing selection, followthe neutral model in evolution. The level of LD was high, the value r2down below0.2atabout2000bp, and no longer down at total4000bp, so there is a close linkage betweenmarkers. There was significant correlations between haplotype (-)Apin-2and three yieldtraits, with contribution value:2.860%、2.901%and2.860%. There were significantcorrelations between2haplotype ((-)Apin-3and (-)Apin-4) and the base yield triat, withcontribution value:1.748%and1.097%. The gene can explain2.86%W,2.86%Wp and5.746%W0, and we thought it was a suitable candidate gene for associated with the yieldtraits.
