Ovotransferrin (ovotransferrin, OVT) expressed in avian oviduct is a class fromtransferrin protein family which can transport iron to the developing embryoglycoprotein,the sequence of OVT have50%homolog with mammalian transferrin andlactoferrin. The researchers found that OVT have a variety of immune activity and hasbeen reported to have anti-viral imune activity as lactoferrin. However, studies on the OVTantiviral mechanism is not very clear. Therefore, this experient use animal experiments andin vitro experiments to study of the antiviral activity and mechanism of OVT, the mainresearch results are as follows:1、 Gavage C57mice with OVT after using VSV virus infect it,then HE stained lungtissue sections. Experimental results show that after OVT gavage, C57mouse lung alveolardamage mitigation, and have a decrease in the number of nucleated cells, so that OVTreduce the VSV virus that causes lung inflammation in mice.2、 OVT incubated with VSV and SeV-infected macrophages C57mice, then usingRT-PCR to detect VSV and SeV virus replication within the cell, the virus titer assay detectVSV replication in cell supernatants situation. Flow cytometry and cell fluorescencestaining detect whether OVT interfere with the binding viruses and cells. The resultsshowed that OVT can inhibit RNA virus replication in macrophages, and this function isnot relate to the interfere of virus binding to macrophages.3、Type I interferon infected macrophages secreted by RT-PCR and Western-blotdetection leads to the secretion of type I interferon transcription factors. Use defectivemouse cell detect type I interferon secretion pathways, and RT-PCR, ChIP study of themechanism of secretion of type I interferons. The results showed that OVT inhibit the geneexpression by inhibiting H3K4and promoting H3K27trimethyl, thereby inhibiting theexpression RNF125, RNF125negatively regulates the RIG-I molecules, which canpromote the expression of RIG-I protein secretion. Therefore OVT through RIG-I/IRF-3pathway to cause the type I interferon secretion and thus play its antiviral activity.
