Ricin is a plant glycoprotein that from castor (Ricinus communis L.) an euphorbiaceae plant, it is oneof the ribosome inactivating protein (RIP). Ricin has RNA N-glycosidase activity, which can kill zooblastespecially some cancer cells. Ricin has the superior research value in the aspects such as antineoplastic,anti-HIV and biopesticide, which has became one of the international research hot spots.This study aimed to discuss the separation and purification technology of ricin, research of ricin asstorage protein and its thermostability and anticancer activity. The main research contents and results are asfollows:(1) The separation and purification of ricin: Castor seed is the experimental material. The total proteinswere extracted by aqueous solution, deposited and concentrated by acetone. We get the crude products bydialysis of the total proteins, and achieve the purification of ricin using affinity chromatography technology.The protein is certified as a purified protein by SDS-PAGE electrophoresis, and proved as the ricin by thelatter cell tests. The innovation of this study lies in the acetone this protein denaturant for processing, notonly realized the protein separation and enrichment, and the extraction of castor oil well, avoid the impactof castor oil for experiments.(2) The research of ricin as storage protein: We analyzed the variation of ricin in the castor seeds usingSDS-PAGE electrophoresis in the process of maturation and germination. Experimental results showed thatthe amount of ricin and the proportion of total proteins in the processed of maturation and germination havethe processes of increase and decrease, namely the ricin showed the behaviors of accumulation andconsumption, thus prove ricin is one of the storage proteins in castor seed. (3) The research of the thermal stability of ricin: We treated the crude ricin with MgCl2, SDS, EDTA,and lacked a control group by water. The degradation situation of ricin is analyzed by SDS-PAGEelectrophoresis analysis. The results displayed that ricin is very stable. The result of ricin as antitumor drugdevelopment provides a strong support.(4) The research of the antitumor activity of ricin: This study inspected the killing effect of ricin on theliver cancer cells and esophageal cancer cells. The experimental results showed that the ricin has significantkilling effect on these cancer cells, ricin is a broad spectrum anticancer drug.
