In this experienment, Tree peony cultivars Paeonia ostii ‘Fengdan’ and peony cultivarsPaeonia lactiflora ‘Fenyunu’ were used as experimental plant material. The study determinated thestorage and viability of pollen,as well as scanning by electron microscopy, simultaneouslycompared with the hybrid parent’s pollen viability changes and pollen morphogical differencesduring storage.The experinment observed and added up the flowering habit and stigmareceptivity,accordingly determined the best pollination period;Meantime,the study explored thecause of the distant hybridization infertility,according to following and observing the stigma andembryos after pollination.In addition,the study also bring up Mature Embryo,and researched theEffect on the Vitro embryo tissue culture of Paeonia ostii ‘Fengdan’ by the Root length anddifferent GA-3and6-BA. Detailed results are as follows:1. Under the same storage conditions,The Paternal Paeonia ostii ‘Fengdan’’s viability ofpollen are all higher than the Female peony cultivars Paeonia lactiflora ‘Fenyunu’’s in differenttimes.With increasing storage time,the pollen viability decreased accordingly,however, theviability of pollen reach to66.03%which are in line with the standard cross-pollination.Theresults of scanning by electron microscopy showed that morphology of pollen are similar to theparental,only differences in the surface ornamentation,and the pollen malformation rate of thePaternal are less than the female’s.2. The Groups of flowering of peony cultivars Paeonia lactiflora ‘Fenyunu’ is25,April to3,May,and28,April into the flowering open period;The process of the length of time such as theslackening bud period,the big bud period to the flowering period generally requires1-2d.Theflowering open period continued4-5d; The stigma begins to secrete mucus on the next day offlowering open period.By tracking observing stigma,the study can determine2-6d is pollinatableperiod.The strongest pollinated period is4-5d that pollen can stick to mucus.3. The distant hybridization between tree peony cultivars and peony cultivars exist mainlyobstacles before fertilization. The pollen of Paeonia ostii ‘Fengdan’ can adhered to penoy stigmaand then germinated.However,the time of germination is delayed. The growth of pollen tube washindered and appeared different forms deformity.Furthermore,That can induce stigma a series ofcallose response.Finally,they can not make pollen tube through the style into the ovary and Complete fertilization.4. There are not generated embryo or embryo abortion in sac through the observation ofovules’ Slice.There are apparent differences between the unit uncompleted double fertilization andthe unit normal natural solid in14d after pollination. Specific performance such as, carpel, surfaceshrinkage, dull.After pollened,the Ovules start to stop increasing and shrinking,then witheredgradually. The experiment finally get49hybrid seeds. The seed rate of distant hybridization is1.414%.5. The embryo tissue culture test shows that the shortest time of epicotyl dormancy breakingis root length at4-5cm through adding hormones.But in that situation,the finnal seedling browningand callusen is more serious.According to ballence the the indicators,we can determine that thesuitable root length is3-4cm so as to breaking epicotyl dormancy.Different seedling with Solublesugar content was not significant. With the increase of root length, Soluble starch content showsdownward trend,and soluble protein content was "down-up-down" trend.6. The two kinds of means such as Only GA3,or GA3and6-BA are all added can all breakepicotyl dormancy.This two kinds of means can increase the speed of breaking epicotyl dormancyand meantime develop the seedling rate.The ratio in shortest time of breaking epicotyl dormancyis6-BA0.1mg·L-1+GA31.0mg·L-1.The best ratio is6-BA0.5mg·L-1+GA31.0mg·L-1.Inaddition,the seedling appeared seedling browning and callusen at the higher concentrations of bothhormones.
