| Porcine Circovirus type2(PCV2) is the major factors for PostweaningMultisystemic Wasting Syndrome (PMWS). Porcine infected with the virus can leadto immune suppression, increased susceptibility to various pathogens. PorcineParvovirus (PPV) and Japanese encephalitis virus (JEV) can cause reproductivedisorders and the boar with performance degradation and other diseases. PCV2andPPV and JEV co-infected pigs had brought enormous economic losses to thecultivation industry. So breeding Pigs are need these three vaccine immunization. Butnow domestic and overseas markets did not have triple vaccine for these diseaseswhich has caused great inconvenience for swine vaccination. In order to effectivelyprevent and control these three diseases, simplify immunization program, reduce theimmune stress, achieve the goal of one immune to prevent many diseases. In thisstudy, we use PCV2XXX strain, PPV XXX strain and JEV XXX strain, which wasisolated and identified by our laboratory, to explore porcine circovirus2typedisease-porcine parvovirus disease-Japanese encephalitis disease trivalent inactivatedtrivalent vaccine. The details are as follows:1. The preparation of inactivated vaccinePCV2with fluorescence quantitative copy number of XXX and PPV withhemagglutinating antibody (HA) of XXX and JEV with TCID50of XXX wereharvested by infecting PK-15cells and BHK-21cells respectively. Admix three kindsof inactivated virus solution which had been XXX times enriched by dialysis withXXX volumes, then append a certain proportion of adjuvant and emulsionize them byorganization stamp mill, PCV2-PPV-JEV triple inactivated vaccines were preparatedsuccessfully. XXX fold diluted concentrated virus solution, mixed XXX volumes ofeach of the two, and three kinds of duplex inactivated vaccine were prepared by usingthe same method; then XXX fold diluted concentrated virus solution and preparedthree kinds of single inactivated vaccines.Levels of alloantigens in the same volumeof inactivated vaccine have the same content. Lay the foundation for the comparisonof the effect of different immunization dose of duplex and triple inactivated vaccine.2. The test of trivalent inactivated vaccine Physical properties and bacterial test of the inactivated vaccine were in line withthe national veterinary biological product quality standards. The test animals (KMmice, guinea pig, piglet, gilts and pregnant sows) showed that no abnormal reactionafter inoculate with the vaccine and pregnant sows showed no abortion, farrowing ratewas100%, the average number of litter was11.4, ratio of weak newborn piglets was1.2%, no records of mummified. The security of inactivated vaccine immunizeexperimental animals and swine is good. Reproductive performance of pregnant sowsand newborn piglets health conditions had no effected, indicating that the vaccine issafe and reliable.3. The immune efficacy test of trivalent inactivated vaccine in35-day-old pigsIn this study, the immunogenicity of the duplex and triple inactivated vaccinecompared with the single and commodity inactivated vaccine shows that, theeffectiveness of different doses of the of the duplex and triple inactivated vaccinewere higher than the single commodity inactivated/attenuated vaccines and salinecontrol group, and can produce high levels of antibodies at low doses (1ml/per head)after immunization. When immune with the same dosage, antibody levels of theduplex and triple inactivated vaccine induced higher than each single inactivatedvaccine, even individual low-dose duplex and triple inactivated vaccine producedslightly higher antibody levels than the single inactivated vaccine. Seven kinds ofantibodies induced by inactivated vaccine can maintain more than10weeks.This study provides the possibility of Porcine Circovirus type2, PorcinePseudorabies virus and Japanese encephalitis virus joint immunization. The studyconfirmed that triple inactivated vaccine induced equivalent or higher antibody thancommodity inactivated vaccine with a low dosage (1ml/per head) immune pigs. |
