| Using male sterile line to produce hybrid seed is one of the main ways of utilization ofHeterosis in wheat. The K-type male sterile lines of wheat are easy to maintain and restore,with having rich restorers and no producing wrinkled seed, etc. Therefore, it is considered tobe the most useful type of sterile lines. In this study, three male sterile lines with AegilopsKotschyi cytoplasm, viz. K3315A, KTP3315A, KTM3315A and72restorer lines of K-typemale sterile wheat as test materials. The1B/1R karyotype of all of the restorer lines wasidentified.3sterile lines were crossed to72restorer lines. Comparing the difference of easyrestoration of fertility in sterile lines of homogeneous heterokaryon and restoring degrees indifferent restorer lines. At the same time, the effects analysis of the restorer genes combinewith molecular markers that tightly linked to restorer genes. The following results wereobtained:1. In order to distinguish1BL/1RS type and non1BL/1RS type of wheat, selectedspecific markers which located on1RS of rye and1BS of wheat for all restorer lines wereamplified by PCR electrophoresis analysis, at the same time, using the A-PAGE method todetect the Gliadin. The results showed karyotype identification results of the two methods areconsistent. In72restorer lines, there are68species belong to the non1BL/1RS type,accounting for94.4%,4restorer lines belong to1BL/1RS type, accounting for5.6%.2. F1seed setting rate of different combinations of72restorer lines and3sterile linesshowed significant difference, the hybrid F1seed setting is widely distributed in the0-91.7%.The same restorer cross with different sterile lines, the easy restoration of fertility amongsterile lines exhibit obvious differences, the order of the easy restoration of fertility in3sterilelines is KTP3315A, KTM3315A, K3315A. Different restorer lines cross with the same sterilelines, and the restoring degrees of different restorers also showed a greater difference.Get2restorer lines with high restoring degrees, such as KR8-1-2(80.80±1.41), KR7-7(80.91±3.73);obtain22stable sterility restorer, such as KR8-1-2(80.80±1.41),KR5212(78.02±2.63),6-96Z6(75.35±1.95),N9806(66.87±1.67)and so on.3. In the72restorer lines, only XY22-2, SY3-1,906E2do not contain fertility restorationgene Rfv1, maybe they have other fertility restoration main genes, such as SY3-1(63.41±19.17). The restorability of906E2(10.41±4.66) is poor, and the effect of restoration gene Rfv2is about10%. The rest of the restorer lines contain the fertility restoration gene Rfv1andRfv2, and the effect of restoration gene Rfv1is approximately60%, the effect of Rfv1isstronger than the effect of Rfv2. The numbers of fertility restoring genes and its effect indifferent restorer lines have yet to be further research. |
