| Populus deltoids has rich germplasm resources, with the quality of straight stem and theyare the most value ones of planting trees in Sect.Aigeiros Duby. Since1970s, they have beenintroduced into China,which promoted the breeding of Poplar. And a series of excellentclones were developed by cross-breeding, but the introduced clones were a few, whichresulted the cross-breeding clones had the similar morphology. This affects the forestryproduction,especially in the fine breed promoted. In order to protect and make full use ofthese clones, it is necessary to use molecular marker to identify the varieties or clones andconstruct their fingerprint. In this study, both SRAP and EST-SSR makers were used toanalysis genetic difference of5Populus deltoides varieties including P.deltoides.cv.‘Lux’(I-69/55) and4varieties related to it, and compared2markers’ analyticalability in the aspect of genetic difference. The results are as follow:1.17polymorphic primers were selected from22primers,86bands were amplificatedwith69polymorphic bands, and polymorphic rate of the bands was80.2%;24specific bandswere amplified, and specific rate of the bands was27.9%.Each pair of primers amplificatedbands among2to10, and5.06bands on average, bands size ranged100bp to450bp.2.21pairs of primers were screened from72primer combinations that could producepolymorphic and high quality bands and identify5varieties completely.287bands wereamplified and209bands were polymorphic from these21pairs of primers and the percentageof polymorphic bands was72.8%.61specific bands were amplified, and specific rate of thebands was21.3%. Each pair of primers amplified13.7bands on average, bands size ranged100bp to1000bp.3. The similarity coefficients of5Populus varieties ranged from0.4444to0.7172with amean of0.5808and its changed in the magnitude was0.2728analysised by EST-SSR; thesimilarity coefficients of5Populus varieties ranged from0.5481to0.7692with a mean of0.6587and its changed in the magnitude was0.2211analysised by SRAP.4.69polymorphic bands amplified by17EST-SSR primers were used for constructingthe EST-SSR unique fingerprint map in5varieties.61specific bands amplified by21SRAPprimer combinations were used for constructing the SRAP unique fingerprint map in5varieties. 5. Compared with EST-SSR,the similarity coefficients of5Populus varieties changed inthe magnitude was smaller analysised by SRAP. Cluster analysis showed that there wasdifference between the genetic similarity assessed by SRAP and EST-SSR markers, whichwas that P. deltoides. cl.‘SL-3’ classified as different group in2markers,but the results ofSRAP and the combination data were consistent. The correlation of the genetic similaritymatrix among the SRAP, EST-SSR and the combination data were analysised, the result werethat the correlation between SRAP and the combination data, EST-SSR and the combinationdata were significant. |
