Bombyx mori cytoplasmic polyhedrosis virus(BmCPV)infects the epithelial cells of themidgut in silkworm larva, leading to midgutpolyhedrosis. The research for pathologicalmechanism of BmCPV can contribute to formulate scientific prevention strategies, andeffectively reduce damage by diseases in the sericultural production. Autophagy is adegradation pathway and participates in multiplicate physiological responses. When cell isinfected virus in mammal, autophagy always plays an important role. In my experiment, wefirst fed mulberry leaf which painted BmCPV for silkworm larver, to making disease model.And then, we objected the expression of ATG8protein using immunohistochemical method,measured the contents of adenosine phosphate utilizing high performance liquidchromatography(HPLC) and detected the expression of BmFoxO genes applying Real-timePCR(qRT-PCR) for the midgut of silkworm larver. To objective, linking BmCPV infected andautophagy in silkworm through autophagy occurs and autophagy induced which involve toAMPK and FoxO signal channel. In the cell save itself aspects, we can explore the controlmeasures when infect BmCPV. Further, we can provide the more theory basis for themechanism of virus generation and precaution. The results are as follows:1The expression of ATG8protein in midgut after the silkworm infected BmCPVWe can obviously watch the pathological phenomena in midgut on62h, when thesilkworm larver fed mulberry leaf which painted BmCPV. The results showed that when thesilkworm infected BmCPV, the expression of ATG8protein was significantly higher thancontrol level within1h to8h, but decreased to background level in8h to35h. Then it sharplyincreased in51h and achieved a peak at99h. The columnar cell of midgut epithelial layerbegan to generate polynedron after infected62h.2The content of ATP,AMP and the ratio of AMP/ATP in midgut after the silkworminfected BmCPVAfter ATP and AMP was measured when the silkworm larver infected BmCPV in midgut,we obtained this result that the content of ATP was significantly higher than control level in 99h and123h. In addition, the content of AMP was significantly higher than control level in1h and35h, but lower than control in51h. Furthermore, to analyze the ratio of AMP/ATP inmidgut of silkworm larver, it was significantly higher than control level, but lower than that in51h、62h、99h and123h.3The expression of BmFoxO gene in midgut after the silkworm infected BmCPVThe expression of BmFoxO gene was analyzed through qRT-PCR method. We haveoutcome that the expression of was BmFoxO gene was significantly lower than control levelin1h and8h in midgut of silkworm larver. However, its expression was unremarkable afterinfected12h.Through my experiment, we initially get the following conclusion that BmCPV infectedin silkworm promoted autophagy activity in midgut during the early infection, but autophagyhelp for that the polynedron was produced and reproduction. At the same time, the AMPKpathway and BmFoxO gene may play a critical role for autophagy induced during the early ofvirus infected.
