Somatic cell cloning technology has great value in theory and in practice. Butgenerally its efficiency is really low and need to be improved. In order to improve theefficiency of somatic cell cloning, researchers have done a lot of work vary from cloneoperation to in vitro culture system to optimize the technology. Among them, mediumoptimization is one of the hotspots.As an antioxidant,esveratrol can regulate cell growth and apoptosis. Epidermal growthfactor (EGF) and insulin like growth factor (IGF) have been shown playing important roleson bovine embryo development. Transferrin and mineral elements are also required forembryonic development. At the same time, many reprogramme promoting factors are addedto the embryo in vitro culture system. In this study, we used EGF, ITS as serum substituteand added resveratrol in traditional medium in order to gain a new components definedmedium. The main contents and results are as follows:1. The parthenogenesis embryo and in vitro fertilization embryo were used as modelsto explore the effects of resveratrol on the in vitro viabilities of bovine embryos. It showsthat0.5uM is the most suitable concentration for embryonic development because of itsaccelerating the embryo cleavage, promoting the blastocyst formation, increasing the cellnumber of blastocyst and the proportion of inner cell mass in blastocysts. Also resveratrolcan reduce apoptosis of blastocyst cells through upregulating apoptosis inhibiting geneBcl-2and downregulating of apoptosis promoting gene Bax. Decrease of ROS in2-cell and4-cell has also been found under resveratrol, which improves embryo anti-oxidative stressand enhance the vitality of embryos.2. Adding EGF, ITS and resveratrol into basic traditional medium (mSOF) instead ofBSA can stably support embryo development to the stage of blastocyst, with the stability ofcell numbers and inner cell mass proportion and the decline of apoptosis rate of blastocystcells.
