Avian paramyxovirus (APMV) belongs to the genus Avulavirus in the family of Paramyxoviridae.The APMVs have been divided into nine different serotypes based on hemagglutination inhibition (HI) and neuraminadase inhibition (NI) assays. APMV has approximately a15kb RNA genome encoding six proteins including hemagglutinin-neuraminidase protein (HN), fusion protein (F), necleocapsid protein (NP), matrix protein (M), phosphoprotein (P), and RNA-directed RNA polymerase(L).NDV (Newcastle Disease virus, NDV) is the only member of the avian paramyxovirus type1. Newcastle disease (ND) is one of the most serious avian diseases. ND can cause substantial economic losses and remains a major threat to the poultry industry worldwide. Low-virulence Newcastle disease viruses (loNDV) are frequently recovered from wild bird species, but little is known about their distribution, genetic diversity, or potential to cause disease in poultry. Therefore, the molecular epidemiology of Avian paramyxovirus bird monitoring is necessary.1Isolation and identification newcastle disease virus from wild birds in the region of XiangHai. In this study, twenty-two NDVs were isolated and identified from shoveller, common goldeneye, gadwall, spot-billed duck respectively. The result showed that the overall separation rate was15.8%. The results showed that all NDV isolates F gene ORF length of1662bp, encoding553amino acid homology of98%-100%. Phylogenetic analysis of partial F gene (47-420bp) showed that22NDVs belong to class I genotype4, having the closest genetic relationship with NDV from JiangSu, Waterfowl and Shorebirds of The United States. Deduced amino acid sequences of the cleavage sites of fusion (F) protein revealed that all strains had the lentogenic molecular characteristics, sharing same motifs112ERQE/GRL117.2The entire genomic sequence of NDV strain Shoveler/JL/374/2012. In order to obtain the complete genome sequence of NDV Shoveler/JL/374/2012,14Pairs of primers were designed to amplify the specific gene fragment by RT-PCR and sequenced. Cloning and sequencing results showed that isolates the whole genome length is15198bp, the same as the whole genome length JS10,9a3b, DE-R49/99and other strains. There was a12-nt insertion (TGGGAGACGGGG) in the phosphoprotein gene between nucleotides2381and2382of the genome (numbered according to the15,186-nucleotide-long genomic sequence of the La Sota strain).3Isolation and identification of avian paramyxovirus serotype6and the HN, F genes sequence analysis. In October2011,300tracheal and cloacal swab samples for wild ducks were collected from XiangHai natural reserve in JiLin province. Eighteen APMV-6isolates were collected from surveillance Program. We amplified, cloned and analyzed the full-length sequence of five strains APMV-6HA and F genes. The five full-length of APMV-6HA and F genes were all1836bp and2031bp respectively. The nucleotide homology were all above99%and shared98%nucleotide identity with TW. The aligning analysis results showed that the five isolates were placed into Class Iof APMV-6.
