Chicken Coccidiosis Disease is one of the modt serious infectious disease which is harmful to poultry. The using of anticoccidial drugs affect the export of poultry products, caused huge economic losses to our country. Egg Yolk antibody(IgY) is an immunoglobulin, using it to replace anticoccidial drugs can overcome many defects. E.tenella is the most common type, which has high pathogenicity and low immunogenicity. We use E.tenella immune hens to collect IgY, and establish the indirect ELISA of E.tenella, providing technical support for the rapid detection.In oral, antibodies is easy enzymatic degradation or loss of activity by stomach acid and pepsin. We use microcapsule technology to protect the antibody resisting the impact of the stomach environment,and release in the intestine.The results were as follows,Verified by sensitivity tests,the indirect ELISA still has a sensitivity after the sera is diluted by1:1600.The recovery rate of IgY was53%through extracted by dilution with water and bitterness-cold ethanol. Through affinity chromatography to further purified, the content of specificity IgY was1.13mg/ml,the pure is82%.Establish the Preparation process of microcapsule:Through the BSA microcapsules to explore the best materials ratio.The result were as follows, the sodium alginate,chitosan,CaCl2optimum conditions were2.0%(w/v),0.2%(w/v),0.5~1.0%(w/v). The drug and resistant starch and sodium alginate proportion for1:2:4. Using the above-described process for preparing antibody microcapsules, the results showed that the capsule drug loading was23.02%, the encapsulation efficiency was75.01%, the protection rate was75.59%,in vitro tests, the release rate of (2h) was less than10%, in the gastric environment,72.14%was intestinal environment release (4h). Meet the requirements of parenteral administration.
