Vitamin E (VE) plays an important role in keeping healthy of human body and promoting reproductive capacity. Therefore, increasing VE content and its activity in Brassica oilseeds advances the nutritive value and quality. We applied ethyl methyl sulfone (EMS) to induce mutagenesis based on the cultivar "ZheShuang72", and a number of mutants with high VE content were identified. Meanwhile, we overexpressed BnaA.VET4.al gene in "ZheShuang72", and acquired four transgenic lines which carry the overexpression gene cassette. Abiotic stress analysis of the four transgenic lines demonstrated the positive function of VE in stress tolerance in rapeseed9Brassic napus). The major findings of this study includes the following1. A feasible and effective method for detection of tocopherol in oilseed rape (Brassica napus L.) using gas ghromatography (GC) with flame ionization detector was found.2. The Tocopherol content and composition of EMS-induced M2population seeds were analyzed.3. DNA polymorphism of genes related to vitamin E synthesis in the EMS-induced M2population by using HRM curve analysis technique was investigated.4. Transgenic lines overexpressing BnaA. VET4.al with "Zheshaung72" background were obtained.5. The transgenic lines were tested for their tolerance against abiotic stresses such as salt (150mM NaCl) and cadmium (100μMCdCl2).
