| To research the distribution and expression of porcine reproductive and respiratorysyndrome virus (PRRSV) receptors in the immune organs of piglets, piglets infected withPRRSV were used to examin pathological changes, immunofluorescence and western blotmethod were used to detect the distribution of virus and receptors, detect the relativeexpression of PRRSV receptors respectively. The results are described as follows:1. There were largely damages in the immune organs of infected piglets with atrophyingof thymus and general swelling of lymphonodi.2. The pathological examination of the infected piglets showed that there was a decreaseof lymphold nodule in the lymphonodi, amygdala and spleen, while the germinal centerdiminished at the same time. The results of immunochemistry SP method revealed that thePRRSV existed in the immune organs of the infected piglets, which indicted that PRRSV hada distinct lymph tissue tropism in piglets.3. The immunofluorescence histochemical staining showed that vimentin was not seen inspleen while Sn, CD163, NMHCⅡ-A all could be found in the immune organs of the infectedpiglets.The distribution of four receptors in the immune organs was more concentrated in thecortex, while more scattered in medulla.4. The results of WB revealed that the expression of the four receptors was significantdifference between congtol group and infected group in immune organs. Comprised tocontrol group,The relative expression of infected group presents an increase in Sn andvimentin and a decrease in NMHC II-A as a whole. The expression of CD163was fluctuatedbetween the conntol and infected group.In summary, porcine reproductive and respiratory syndrome virus mainly damageimmune organs of piglets, and results in immunosuppression and persistent infection, whichsuggested that the four receptos has the synergistic effects, while every receptor has differenteffect on different tissue in the process of PRRSV infection. |
